Description of ATN-161 trifluoroacetate salt (ATN-161 TFA salt)

ATN-161 trifluoroacetate salt (ATN-161 TFA salt) is a non-RGD fibronectin-derived pentapeptide integrin α5β1/αVβ3/αVβ5 antagonist. ATN-161 trifluoroacetate salt blocks fibronectin binding and αVβ5 interaction with ACE2/SARS-CoV-2 spike protein to inhibit endothelial migration, angiogenesis, and viral entry into host cells. ATN-161 trifluoroacetate salt reduces tumor metastasis and inflammation by interfering with integrin-mediated adhesion and Rho/NF-κB signaling. ATN-161 trifluoroacetate salt is useful for research on solid tumors and metastasis, choroidal neovascularization, ischemic stroke, and COVID-19[1-4].

In vitro, treatment of TSC2-deficient lung vascular smooth muscle cells with 10μM ATN-161 trifluoroacetate salt for 4 days blocked matrix-induced cell proliferation[5]. Treatment of pluripotent stem cell-derived hematopoietic differentiation cells with 10μM ATN-161 trifluoroacetate salt for 2-6 days did not affect RUNX1, GATA2, and MYB expression. Treatment of pluripotent stem cell-derived hematopoietic differentiation cells with 10μM ATN-161 trifluoroacetate salt for 2-6 days did not affect CD43+ HPCs generation[6]. Pretreatment of periodontal ligament stem cells with 10μM ATN-161 trifluoroacetate salt for 24 hours reduced Porphyromonas gingivalis infection in periodontal ligament stem cells[7].

In vivo, tail vein injection of 30μg/mouse ATN-161 trifluoroacetate salt twice per week for 5 weeks in NSG mice orthotopically injected with LM2 cells inhibited primary tumor growth, whereas​ ATN-161 trifluoroacetate salt did not reduce spontaneous lung metastasis[8]. A single intravitreal injection of 5μL 0.1-10μg/mL ATN-161 trifluoroacetate salt in Brown-Norway rats after laser-induced choroidal neovascularization inhibited choroidal neovascularization leakage and neovascularization. A single intravitreal injection of 5μL 0.1-10μg/mL ATN-161 trifluoroacetate salt in Brown-Norway rats after laser-induced choroidal neovascularization reduced choroidal neovascularization area and thickness, decreased VEGF expression in the RPE-choroid complex[9]. Intraperitoneal injection of 100mg/kg ATN-161 trifluoroacetate salt every 3 days for 21 days starting from day 4 after intrasplenic injection of CT26 cells in BALB/c mice inhibited liver metastasis tumor angiogenesis. Combination with 100mg/kg 5-FU infusion reduced liver metastasis number and liver weight, decreased tumor cell proliferation, increased tumor cell apoptosis, and improved overall survival[10].

Human periodontal ligament stem cells (PDLSCs) and human oral keratinocytes (HOK)

Preparation Method

PDLSCs and HOK were pretreated with 10μM ATN-161 trifluoroacetate salt for 24 hours, followed by infection with P. gingivalis ATCC 33277 at MOI 100 for 2 hours, then cells were washed and lysed to release intracellular bacteria for CFU counting, or fixed for immunofluorescence staining.

ATN-161 trifluoroacetate salt reduced P. gingivalis infection in PDLSCs, but showed poor inhibitory effect on P. gingivalis infection in HOK.

Preparation Method

Mice were orthotopically injected with 104 LM2 cells into mammary fat pad. When tumors reached 2mm in diameter, mice were intravenously treated with ATN-161 trifluoroacetate salt (30μg/mouse) twice per week for 5 weeks. Primary tumors and lungs were collected for Western blotting, immunohistochemistry staining, and bioluminescence imaging.

Applications

ATN-161 trifluoroacetate salt suppressed focal adhesion kinase activation and tumor cell proliferation in primary tumors, but ATN-161 trifluoroacetate salt did not reduce spontaneous lung metastasis or inhibit tumor cell proliferation in lung metastatic nodules.
