Fluorescent probes that bind with calcium ions and exhibit spectral reactions allow researchers to observe changes in intracellular free calcium ion levels, which can be detected through fluorescence microscopy, flow cytometry, fluorescence spectrophotometer, and fluorescence enzyme-linked immunosorbent assay (ELISA) plates.

The new generation of calcium ion fluorescence probes: Cal-520, Ca-590, and Cal-630 are the strongest and most uniform fluorescent probes for measuring intracellular calcium mobility. Compared with traditional calcium ion probes such as Fluo-3AM, Fluo-4 AM, and Rhod-2 AM, they have significantly improved signal-to-noise ratio and intracellular retention

Cal-520 AM, Ca-590 AM, and Cal-630 AM have cell permeability and can be preloaded into cells. Once inside the cell, the lipophilic blocking group AM is cleaved by esterase, producing negatively charged fluorescent dyes that remain inside the cell. Once combined with calcium ions, their fluorescence is significantly enhanced. When cells are stimulated by agonists, receptors emit signals of intracellular calcium release, thereby increasing the fluorescence of Cal-520, Cal-590, and Cal-630. The high sensitivity and>100 fold fluorescence enhancement of Cal-520 AM, Cal-590 AM, and Cal-630 AM make them ideal probes for intracellular ion determination. High signal-to-noise ratio and greater intracellular retention make it an excellent tool for evaluating GPCR and calcium channel targets, as well as screening corresponding agonists and antagonists.

Cal-520, Ca1-590, and Cal-630 are mainly located in the cytoplasm, unlike Rhod-2, which is mainly located in mitochondria. The long excitation/emission wavelengths of Cal-590 and Cal-630 make them perfect calcium ion probes and compatible with green fluorescent white (GFP) labeled cells for multicolor detection. In addition, Cal-520, Cal-590, and Cal-630 are also compatible with the vast majority of existing optical instruments. Cal-520 was well excited at 488nm and detected using a FlTC filter; Csl-590 was optimized to be well excited outside 555nm and detected using TRITC/Cv3 filters. Cal-630 was optimized to be well excited outside 594nm and detected using Texas Red filters. Cal-590 Potassium Salt is a potassium form of Ca-590, which does not have fine permeability and is soluble in water. It can be loaded into cells through microinjection, membrane electrodes, scratch marks, and other methods to detect intracellular calcium levels.
