Protocol of D-Sorbitol-13C6

Ce protocole décrit l'utilisation de D-Sorbitol-13C6 pour le traçage du flux métabolique de l'utilisation et du transport du sucre dans les ostéoclastes résorbants l'os. Les conditions d'application réelles peuvent nécessiter une optimisation en fonction du type de cellule et de la dose du traceur[1–2].

1.Préparation des Réactifs et Matériels

D-Sorbitol-13C6, des ostéoclastes matures dérivés de macrophages de la moelle murins (BMMs), M-CSF (30ng/mL), RANKL (50ng/mL), α-MEM sans glucose supplémenté avec 10% de FBS dialysé, LC-MS/MS avec des étalons internes appropriés.

2.Etapes Expérimentales

(1)Différenciation des Ostéoclastes

Isoler BMMs de rat et les différencier dans α-MEM avec M-CSF et RANKL pendant 5 jours jusqu'à ce que des ostéoclastes matures se forment (cellules multinucléées TRAP+ ). Passer à un milieu sans glucose 6-12 heures avant le marquage pour réduire la réserve de carbone non-marqué en arrière-plan.

(2)Traitement au Traceur

Préparer 1-5mM de D-Sorbitol-13C6 dans α-MEM sans glucose avec 10% de FBS dialysé. Ajouter le milieu traceur aux puits (500µL-1mL par puits) et incuber les cellules à 37°C, 5% de CO2 pendant 1-4 heures en fonction de la réssolution de flux requise.

(3)Extraction des Métabolites

Laver vite les cellules avec PBS froid. Arrêter le métabolisme en ajoutant du méthanol froid à 80% (pré-refroidi à -80°C). Râper les cellules et transférer le lysat vers des tubes à microcentrifuger. Centrifuger à 14000rpm, 4°C, pendant 10 minute. Recueillir le surnageant pour l'analyse LC-MS/MS.

(4)Analyse LC-MS/MS

Analyser les métabolites marqués en utilisant LC-MS/MS à triple quadripôle en mode ESI négatif. Suivre les transitions des sucres 13C-marqués et non-marqués (par exemple, glucose, fructose, sorbitol, G6P). Utiliser D-Sorbitol-13C6 comme étalon interne pour la normalisation lors de la quantification de D-sorbitol endogène. Normaliser en fonction de la teneur en protéines ou du nombre de cellules si nécessaire.

Remarques et Précautions

(1)Optimisation de la dose du traceur : des concentrations élevées de sorbitol peuvent affecter l'équilibre osmotique-effectuer une titration pour trouver une dose non-toxique.

(2)Conception du suivi temporel : Pour l'analyse dynamique de flux, collecter des échantillons à plusieurs points temporels (par exemple, 0, 1, 2, 4h).

(3)Chevaucher spectral de masse: S'assurer que les formes marquées et non-marquées peuvent être résolues au niveau de la ligne de base.

(4)Témoins : Inclure un témoin de D-sorbitol non-marqué et un blanc "sans traceur" pour la correction du fond.

(5)Imagerie en aval : la co-incubation avec LysoTracker ou LAMP2 peut aider à localiser les réserves de sucres marqués.

(6)Stockage : Conserver les échantillons à -80°C jusqu'à l'analyse pour prévenir la dégradation.
