Détermination de l'activité TAFIa dans le plasma par la méthode Hippuryl-Arg-OH[1]
Ce protocol est adapté de données d'étude et frouni à titre de référence uniquement. Des ajustements peuvent être nécessaires selon les exigences expérimentales spécifiques.
1. Préparation des réactifs: Préparer une solution de travail 20mM du substrat Hippuryl-Arg. Préparer des solutions de travail de Thrombine à 5U/mL et de Thrombomoduline à 7.5U/mL, puis les mélanger pour former un complexe thrombine-thrombomoduline. Préparer une solution de CaCl₂ à 60mM. Préparer un tampon d'activation (HEPES 20mM , NaCl 0.15M, CaCl₂ 5mM, pH=7.4). Préparer une solution de HCl 1M pour arrêter la réaction. Préparer une solution de NaOH 1M. Préparer un tampon phosphate 1M. Préparer une solution de chlorure de cyanuryle à 3% dans du 1,4-dioxane.
2. Activation du TAFI : Prélever 15μL d'échantillon de plasma et le mélanger avec 12μL du complexe thrombine-thrombomoduline, 8.5μL de la solution de CaCl₂ 60mM et 24μL de tempon d'activation. Incuber le mélange à température ambiante durant 10 minutes pour activer le TAFI présent dans le plasma en TAFIa. Ensuite, ajouter 20μL d'Hirudine pour arrêter la réaction d'activation.
3. Détection de l'activité TAFIa: Au mélange réactionnel de l'étape précédente, ajouter 20μL de la solution de travail du substrat Hippuryl-Arg 20mM et incuber à température ambiante durant 10 minutes. Puis, ajouter 20μL de la solution de HCI 1M pour arrêter la réaction du substrat. Ensuite, ajouter successivement 20μL de la solution de NaOH 1M et 25μL du tampon phosphate 1M dans le système réactionnel. Enfin, ajouter 60μL de la solution de chlorure de cyanuryle à 3%dans le 1,4-dioxane et laisser reposer pour un développement de couleur suffisant.
4. Détection et calcul: Centrifuger le mélange réactionnel ayant développé sa coleur à 14,000g durant 2 minutes. Prélever le surnageant et mesurer son absorbance à une longeur d'onde de 405nm à l'aide d'un spectrophotomètre. Tous les échantillons et les points de la courbe standard doivent être dilués à 50% avec un sérum inactivé par la chaleur et mesurés en double exemplaire.
