Modified Giemsa Staining Solution(20X)

Instructions for use:

1. Preparation of reagents

Preparation of Improved Giemsa Staining Solution (1X): According to the number of samples, take an appropriate amount of Improved Giemsa Staining Solution (20X), add deionized water or double distilled water to dilute 20 times to obtain Improved Giemsa Staining Solution (1X). For example, 1ml of modified Giemsa staining solution (20X) is added to 19ml of water and mixed evenly to obtain 20ml of modified Giemsa staining working solution (1X). Note: The diluted improved Giemsa staining working solution (1X) is a ready to use reagent and should not be stored. It should be used and prepared immediately to avoid affecting the staining effect.

2. Sample processing

a. For paraffin sections:

Dewaxing in xylene for 5-10 minutes.

Replace with fresh xylene and dewaxing for 5-10 minutes.

Anhydrous ethanol for 5 minutes.

90% ethanol for 2 minutes.

80% ethanol for 2 minutes.

70% ethanol for 2 minutes.

b. For frozen sections:

Wash with distilled water for 2 minutes.

c. For blood or bone marrow smears:

Prepare blood or bone marrow smears using conventional methods and air dry them naturally.

Fix with 70% ethanol for 10 minutes.

d. For cultured cells:

Add 70% ethanol and fix for 10 minutes.

3. Improved Giemsa staining

a. Add an appropriate amount of prepared modified Giemsa staining solution (1X) to the sliced or smeared samples and stain for 45 minutes. Note: If the staining is too deep or too light, the staining time or working solution concentration should be adjusted.

b. Wash thoroughly from one side with distilled water, and after drying, observe and take photos under a microscope.
