Description of AZD7545

AZD7545 es un inhibidor de tipo competitivo del piruvato deshidrogenasa quinasa 2 (PDHK2) (IC₅₀=6.4nM) que también zinhibe la actividad de PDHK1 (IC₅₀=36.8nM)[1]. AZD7545 inhibe la actividad de PDHK al interrumpir la interacción entre PDHK2 y el dominio lipoilo interno del componente dihidrolipoamida acetiltransferasa del complejo de piruvato deshidrogenasa (PDC), aumentando así la proporción de PDH activo[2]. AZD7545 lo usas para estudiar regulación del metaboliusmo de la glucosa[3]. AZD7545 también inhibe el crecimiento de células de melanoma no les afectan los medicamentos[4].

In vitro, el tratamiento de células de cáncer de pulmón de células no pequeñas A549 con AZD7545 (10–20μM) durante 4–24 horas inhibió eficazmente la fosforilación del piruvato deshidrogenasa (PDH) y mejoró su actividad[5]. El tratamiento de células de cáncer de pulmón de células no pequeñas H1299 con AZD7545 (500μM) solo durante 24 horas inhibió la actividad de PDK1 y mejoró la actividad de PDH, pero no indujo significativamente la apoptosis de las células de cáncer; sin embargo, cuando se combina con el activador PKM2 ML265 (500μmol/L), AZD7545 inhibió sinérgicamente la proliferación celular e indujo la apoptosis, al tiempo que redujo el consumo de glucosa y la producción de lactato, promoviendo la remodelación metabólica de la fosforilación oxidativa[6].

In vivo, la administración de AZD7545 (0.5g/L) a través de agua potable durante 5 semanas a ratones C57BL/6J hembra ovariectomizados (OVX) de 7 semanas de edad suprimió significativamente la pérdida ósea inducida por OVX y redujo la activación de la vía de señalización NFATc1 en los osteoclastos[7]. Una dosis oral única de AZD7545 (10–30mg/kg) administrada a ratas Wistar y ratas obesas de Zucker (fa/fa) activó significativamente la piruvato deshidrogenasa (PDH) en el hígado y el músculo esquelético y mejoró el control delo azúcar en sangre en ratas Zucker obesas[8].

Células A549 (línea celular de cáncer de pulmón de células humanas no pequeñas)

Método de preparación

Las células A549 se mantuvieron en un medio RPMI 1640 suplementado con un 10% de suero bovino fetal inactivado por calor (FBS) y un 1% de penicilina/estreptomicina a 37 °C, 5% de CO2. Se añadió 10-20μM AZD7545 durante 2-24h.

Áreas de aplicación

AZD7545 mejoró efectivamente la actividad de PDH en células A549 al reducir las fosforilaciones en los residuos S293 y S300 de PDHA1. El tratamiento con AZD7545 aumentó la tasa de consumo de oxígeno (OCR) y disminuyó la tasa de acidificación extracelular (ECAR) en las células, lo que indica un cambio de la glucólisis a la fosforilación oxidativa.

Modelos animales

Ratas Wistar y ratas obesas Zucker (fa/fa)

Método de preparación

A las ratas Wistar macho (200-240 g) se les administró por vía oral una dosis única de AZD7545 (10-30 mg/kg) suspendida en 0,5% (p/p) de metocel/0,1% de polisorbato 80 a las 08:00h. Las ratas obesas de Zucker fueron tratadas por vía oral con AZD7545 dos veces al día durante 7 días. Los animales fueron sacrificados 2 horas después de la administración de una sola dosis para la recolección de tejidos.

Forma de dosificación

10-30 mg/kg; gavage oral

Áreas de aplicación

Una dosis única de AZD7545 aumentó significativamente la proporción de PDH activo (desfosforilado) en el hígado y en el músculo esquelético en ratas Wistar. En ratas Zucker obesas. El tratamiento con AZD7545 también aumentó la actividad de la PDH muscular del 61,0% al 97,0% PDH activa y hepática en ratas Zucker obesas.
