Biotin-PEG3-azide es la forma PEGified de biotin-azide y se puede utilizar en la síntesis de PROTACs [1]. Biotin-PEG3-azide se puede utilizar para etiquetar el alquino termial a través de una reacción de clic catalizada por cobre o una reacción de clic de descobre con ciclooctina [2]. Biotin-PEG3-azide se puede utilizar en reacciones no enzimáticas y marcado covalente de proteínas, y se puede separar y detectar mediante métodos de laboratorio comunes como SDS-PAGE y Western blotting [3].

Análisis de microscopía de fluorescencia de sondas intracelulares [1]:
1. Coloque las células en una fuente de cultivo de 35 mm recubierta con poli-L-lisina (con una concentración del 1% en DPBS) y permita que crezcan hasta aproximadamente un 50 % de densidad celular.
2. Lave las células tres veces con DPBS y cultúrelas en un medio que contenga 10 μM de la sonda objetivo (10 μL de solución comercial de 1 mM disuelta en DMSO) durante 4 horas (el tiempo de incubación y la concentración se determinan de acuerdo con las instrucciones proporcionadas por el material de la sonda comprado).
3. Después de la incubación, lave las células tres veces con DPBS y incubelas a temperatura ambiente durante 15 minutos en formalina al 4% (en DPBS).
4. Las células fijas se enjuagan a fondo e incuban a temperatura ambiente durante 1 hora con Click Master Mix (Biotin-PEG3-azide 10μM, THPTA 100mM, ascorbato de sodio 100mM y CuSO4 15mM).
5. Las células se lavan tres veces con DPBS, tres veces con solución de cloruro de sodio anhidra de 500 mM y luego se enjuagan con agua desionizada.
6. Las células se incuban en la oscuridad con estreptavidina-A488 (1 ml, preparada diluyendo la solución madre de 1 mg/mL con DPBS 1:1000) durante 30 minutos. Las células se lavan con DPBS y se incuban a temperatura ambiente con DAPI (1 ml, 50 nM disueltos en DPBS) durante 5 minutos. Finalmente, las células se lavan con DPBS y se someten a imágenes de fluorescencia.
