Bufalin es un inhibidor de Na+/K+-ATPase (α1: Kd = 42.5nM; α2: Kd = 45nM; α3: Kd = 40nM) [1]. Bufalin inhibe principalmente la actividad de Na⁺/K⁺-ATPase en las membranas celulares, lo que lleva a concentraciones elevadas de cacio intracelular[2]. La inducción de Bufalin de la proliferación de células tumorales también puede tener múltiples mecanismos antitumorales, incluido el desencadenamiento de la autofagia y la inhibición de la angiogénesis tumoral [3]. Bufalin se utiliza para tratar varios tumores, incluyendo cáncer de hígado, cáncer de pulmón, cáncer de mama y leucemia[4].
En las células NCI-H640, después del tratamiento con Bufalin (1μM, 2μM, 4μM; 24h), la actividad de las células disminuyó significativamente [5]. En las células de cáncer de ovario Ishikawa, SK-OV-3 y OMC-3, Bufalin (0.1-10ng/mL; 48h) puede inhibir la proliferación de células de cáncer de endometrio y de ovario [6]. En las células RCC ACHN humanas, Bufalin (0-80nM; 12-72h) suprime la proliferación y metástasis del carcinoma de células renales al inhibir la vía de señalización PI3K/Akt/mTOR [7].
En el modelo de rata de edema de patas inducido por carragenena, después del tratamiento con Bufalin (0.15mg/kg, 0.3mg/kg, 0.6mg/kg; ip; inyección única), el volumen de edema de patas en ratas se redujo significativamente [8]. En el modelo de ratón de tumor ortotópico metastásico de células HCCLM3, los tejidos tumorales trasplantados ortotópicamente sufren necrosis y apoptosis inducida por el tratamiento con Bufalin (1mg/kg, 1.5mg/kg; ip; 30d) [9].

Método de preparación | Aproximadamente 2 × 105 células/pocillo de células NCI-H640 en placas de 12 pozos se incubaron con Bufalin a concentraciones finales de 0 (vehículo, 0,5 % DMSO), 1, 2 y 4 μM durante 24 horas. Las células de cada tratamiento se tiñeron con PI (5 g/ml) y se analizaron mediante citometría de flujo y se calculó la viabilidad celular. |

Método de preparación | El efecto antiinflamatorio se basó en la inhibición del edema de la pata trasera inducido por carragenina. Se administraron bufalina (0,15, 0,3 y 0,6 mg/kg, i.p.), indometacina (10 mg/kg, i.p.) y vehículo por vía intraperitoneal 30 minutos antes de la inyección de carragena. Cada rata recibió una inyección subplantar de carragenina (0,1 ml, 1% p/v en solución salina) en la pata trasera derecha. Un pletismómetro midió el volumen del edema de la pata antes y en varios momentos (1, 2, 4 y 6h) después de la inyección de carragenina. La proporción de hinchazón de la pata se calculó como un aumento porcentual del volumen de la pata medido antes de la inyección de carragenina. Las patas de rata fueron recogidas después de la evaluación final. Cada pata trasera se cortó a nivel del hueso calcáneo y se utilizó para el análisis de Western blot. |
