cKK-E12 pertenece a los lípidos ionizables. Es capaz de conformar nanopartículas lipídicas, ideales para transportar de forma efectiva el RNA interferente pequeño y el RNA mensajero[1]. Las estructuras lipídicas creadas coneste compuesto tienen una gran capacidad de adherirse a la apolipoproteína E presente en el organismo. Por lo tanto, tienden a localizarse preferentemente en los hepatocitos hepáticos[2]. Se emplea en múltiples ámbitos, desde la admnistración génica dirigida y tratameintos terapéuticos hasta la elaboración de vacunas y térnicas de imagen molecular[3,4].
En cultivos celulares, se expusieron macrófagos RAW 264.7 400ng/mL de nanopartículas lipídicas de cKK-E12 durante seis horas. La intervención elevó de forma considerable la síntesis de citocinas proinflamatorias como IL-6, TNF-α, IL-1α e IFN-β, además del quimiocito MCP-1. Si se prolonga el tratamiento hasta 24 horas con la misma concentración, ccrece la cantidad de células que sufren apoptosis, necrosis y piroptosis[5].
En ensayos de animales, se inyectó por vena caudal nanopartículas lipídicas de cKK-E12 cargadas con RNA mensajero marcado con Cy5 a ratones C57BL/6, con dosis de 0.75mg/kg. TRanscurrida una hora, más del sesenta por ciento del material se acumuló en el hígado. El bazo, pulmones y riñones albergaron el resto del compuesto[6]. Al aplicar por vía intravenosa partículas cargadas con mRNA mensajero de trastuzumab en dosis de 0.5, 1 y 2mg/kg en ejemplares de la misma cepa, el nivel sérico de trastuzumab subió según la dosis suministrada en 24 horas. La concentración llegó a unos 40µg/mL cuando se usó la dosis máxima de 2mg/kg[7].

Método de preparación | Los ratones C57BL/6 fueron inyectados a través de la vena de la cola con cKK-E12 LNP que encapsulaba mRNA marcado con Cy5 (dosis de 0.75mg/kg). 1h después de la inyección, los ratones fueron sacrificados, y los órganos principlaes (hígado, bazo, pulmones y riñones) fueron recogidos y sometidos a imágenes de fluorescencia ex vivo utilizando un sistema de imágenes IVIS (Ex/Em: 649/670nm). La biodistribución porcentual se calculó para cada órgano basándose en la eficiencia radiante total analizada a través de la región de interés utilizando el software Living Image. |
