Dynorphin A (trifluoroacetate salt) es un péptido opioide endógeno que activa el canal receptor huérfano XOR1-K+ , con un valor EC50 de 45nM [1]. Dynorphin A (trifluoroacetate salt) puede unirse y activar a todos los familias de receptores opioides, regulando flujo iónico de la membranan de las neuronas[2]. Dynorphin A (trifluoroacetate salt) se ha aplicado en sestudios relacionados con la necesidad de las drogas ty la adición, así como en la regulación de las funciones neuronales[3].
In vitro, el tratamiento con Dynorphin A (trifluoroacetate salt) a 10μM durante 4 horas indujo la activación de la caspase-3 en las células neuronales estriatales del ratón y aumentó el nivel de citocromo c liberado de mitocondrias[4]. El tratamiento con Dynorphin A (trifluoroacetate salt) (10μM) mientras 16 horas puede provocar una fuete actividad de respeusta de migración en las células HEK293 que expresan Mac-1[5]. El tratamiento con 100μM de Dynorphin A (trifluoroacetate salt) durante 96 horas indujo la muerte propios de las neuronas la médula espinal de ratón [6]. Método de preparación | Las células U2OS se cultivaron en medio DMEM, que se complementó con un 10% de suero bovino fetal (FBS), L-glutamine y 1% de penicillin/streptomycin. Las células fueron cultivadas en condiciones de 37°C y 5% de CO2. Se inocularon 3000 células U2OS en cada pozo de una placa de 96 pozos. Las células se tartaron con Dynorphin A (trifluoroacetate salt) a concentraciones de 0.1, 0.5, 1, 5, 10, 50 y 100μM durante 48 horas. Se añadieron 10μl de reactivo CCK-8 a cada pozo y se incubaron durante 2 horas. Luego, el valor de absorbancia se midió a 450nm. |
