GABA es un neurotransmisor inhibidor endógeno fundamental en el cerebro de los mamíferos. Presenta valores de EC50 de 0.17, 2.1, 2.1 para 1.4μM para α6, α1, α4 y α5 del receptor GABAA[1]. GABA se almacena en vesículas sinápticas gracias a los transportes vesiculares de aminoácidos inhibidores. Regula la excitabilidad neuronal opara mantener el equilibrio de las funciones cerebrales[2]. Se usa mucho en diferentes modelos animales para modular la respuesta inmune, interferir en el desarrollo tumoral y aliviar la ansiedad[3].
En estudios in vitro, el tratamiento con GABA (100µM) durante 72 horas frenó la proliferación de células SW480 y SW620 y HCT116, y provocó la parada del ciclo celular[4]. La exposición de células B de islotes de ratón a 100µM de GABA durante 4 días estimuló su proliferación. Se observó un aumento de la expresión nuclear de p-Akt Ser473 y p-p70S6K Thr421/Ser424[5].
En estudios in vivo, la administración oral de GABA 100mg/kg/día durante las semanas mejoró el estado metabólico de ratas con síndrome de ovario poliquístico (PCOs). También redujo el peso corporal, el índice de masa corporal y los niveles de testosterona[6]. la inyección intraperitoneal diaria de 1.5mg/kg de GABA durante 3 meses mejoró los niveles de glucosa e insulina, el estado metabólico y la grasa corporal en ratas diabéticas[7]. El pretratamiento con una dosis única de GABA (10mg/kg; i.p.) una hora antes aumentó la supervivencia de ratones con lesión pulmonar aguda por LPS. Alvió la lesión inflamatoria y el edema pulmonar, redujo la myeloperoxidase (MPO), bajó las citocinas proinflamatorias IL-1β y TNF-α, y elevó la citocina antiinflamatoria IL-10[8].

Método de preparación | Las células SW480 se cultivaron en el medio de Eagle modificado de Dulbecco (DMEM), suplementado con un 10% de suero de ternera fetal (FBS), y se mantuvieron a 37°C en una atmósfera de aire humidificado con un 5% de CO2. Las células se sembraron a una densidad de 1300 células/pocillo en placas de 96 pozos, que contenían 100μl del medio DMEM. Veinticuatro horas después, las células fueron incubadas con GABA (0, 10, 50 y 100µM) durante 24, 48 y 72h, respectivamente. El ensayo de proliferación celular se realizó utilizando el ensayo CCK-8, y se midieron los valores de densidad óptica a 450nm. |

Modelos animales | Ratones macho C57BL/6 |
Método de preparación | 128 ratones C57BL/6 machos sanos de 6–8 semanas de edad, con un peso de 18-22g, fueron criados en jaulas en un entorno específico libre de patógenos (SPF), con alimentos y agua adecuados, y la temperatura del laboratorio se mantuvo en alrededor de 25°C. 36 ratones fueron tomados y anestesiados por inyección intraperitoneal de 1g/kg de hidrato de cloral al 4%. Los ratones se dividieron en los siguientes 3 grupos, con 12 ratones en cada grupo: grupo control (0.2ml de solución salina normal para inyección intraperitoneal), grupo LPS (30mg/kg de LPS para inyección intraperitoneal), grupo GABA+LPS (10mg/kg GABA se administró por inyección intraperitoneal 1 hora antes de la prueba de LPS). Los ratones fueron anestesiados 6 horas después de LPS o inyección de solución salina, y se recogieron tejidos pulmonares para su análisis. |
