Description of GPX4 16

GPX4 16 is an inhibitor of glutathione peroxidase 4 (GPX4).[1] It inhibits the proliferation of Nthy-ori-3-1 transformed human thyroid follicular epithelial cells (IC50 = 8.39 µM), as well as MDA-T32 and MDA-T41 human thyroid cancer cells (IC50s = 10.29 and 8.18 µM, respectively). GPX4 16 (10 µM) induces ferroptosis in MDA-T32 cells, an effect that can be prevented by the ferroptosis inhibitor ferrostatin-1 . It reduces GPX4 protein levels in MDA-T32 cells when used at a concentration of 0.3 µM. GPX4 16 also induces the production of reactive oxygen species (ROS) and reduces glutathione (GSH) levels, mitochondrial polarization, and mitochondrial respiration in MDA-T32 cells.

Calculation results:

Working concentration: mg/ml;

Method for preparing DMSO master liquid: mg drug pre-dissolved in μL DMSO ( Master liquid concentration mg/mL, Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug. )

Method for preparing in vivo formulation: Take μL DMSO master liquid, next addμL PEG300, mix and clarify, next addμL Tween 80, mix and clarify, next add μL saline, mix and clarify.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next add μL Corn oil, mix and clarify.

Note: 1. Please make sure the liquid is clear before adding the next solvent.
2. Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such as vortex, ultrasound or hot water bath can be used to aid dissolving.
