Description of Matrine

Matrine es un alcaloide que se encuentra en las plantas de Sophora japonica y actúa como agonista de los receptores opioides kappa y µ. La matrina presenta múltiples efectos farmacológicos, incluyendo propiedades anticancerígenas, antioxidantes, antiinflamatorias y antiapoptóticas.

En estudios in vitro, el tratamiento de las células A549, DU145 y MIA PaCa-2 con matrina (50 μM) durante 24 horas inhibió la expresión del receptor tipo 4 de quimioquina CXC (CXCR4), downreguló los niveles de metaloproteinasas de matriz (MMP-2 y MMP-9), y debilitó la actividad de migración celular. En la línea celular de cáncer gástrico MNK45, el tratamiento con matrina (50-500 μg/ml) inhibió la proliferación celular de manera dependiente de la dosis, con un valor de IC50 de 540 μg/ml, y afectó significativamente la expresión de proteínas intracelulares como NF-κB, XIAP, CIAP y p-ERK. Además, el tratamiento de células D341 con matrina (0.5-2.0 mg/mL) durante 24-72 horas indujo apoptosis de manera dependiente de la dosis y aumentó la expresión de genes relacionados con la apoptosis, como Bax, caspasa-3 y caspasa-9.

En estudios in vivo, se administró matrina (300 mg/kg) por vía oral a ratas con cardiomiopatía diabética (DbCM) durante 10 días, lo que redujo el daño a la función del ventrículo izquierdo, restauró la pérdida de compliance cardíaca y inhibió la fibrosis cardíaca causada por la activación de la vía de señalización TGF-β1/Smad. También se inyectó matrina (150 y 25 mg/kg) por vía intraperitoneal a ratas con encefalomielitis autoinmunitaria experimental (EAE), lo que redujo la severidad clínica de la EAE, disminuyó la infiltración celular en el sistema nervioso central, protegió la expresión de colágeno tipo IV y de ARNm de ZO-1, y reguló el equilibrio entre MMP-9 e inhibidor tisular de metaloproteinasas (TIMP-1) en suero.

Método de preparación

Las células fueron tratadas con 50 µM de matrina durante diferentes intervalos de tiempo (0, 6, 12, 24 h). El nivel de ARNm se midió mediante RT-PCR, utilizando GAPDH como control de carga. Luego, se compararon los niveles de expresión de CXCR4, MMP-2 y MMP-9 en cada línea celular.

Condiciones de reacción

Áreas de aplicación

La matrina disminuyó la expresión de CXCR4, así como los niveles de MMP-2 y MMP-9 en las células A549, DU145 y MIA PaCa-2.

Método de preparación

Se utilizó una inyección intraperitoneal de estreptozotocina (65 mg/kg) para inducir diabetes en las ratas. Antes de la inducción de diabetes, las ratas recibieron matrina por vía oral a una dosis de 300 mg/kg por día durante 10 días.

300 mg/kg/día; p.o.

La matrina atenúa el deterioro de las funciones del ventrículo izquierdo en un modelo experimental de cardiomiopatía diabética (DbCM), recupera la pérdida de compliance cardíaca e inhibe la fibrosis cardíaca inducida por la activación de la vía de señalización TGF-β1/Smad.
