Description of ME1111

ME1111 es un nuevo antimicótico de uso tópico[1-2]. Cuenta con gran capacidad para penetrar la uña, por lo que llega sin dificultad a las zonas infectadas y frena el desarrollo de hongos patógenos, especialmente los dermatofitos. Se emplea en estudios sobre la onicomicosis y otras afecciones por hongos[3-4].

Se expusieron las líneas celulares humanas K562, HepG2, U937 y A431 a ME1111 (0.06–32μg/mL) durante 72 horas. Los valores de IC50 registrados oscilaron entre 33 y 47μg/mL en estas cuatro cepas celulares[5]. Además, se tratan cepas de Trichophyton rubrum strains y Trichophyton mentagrophytes strains (0.004–8μg/mL) durante 96 horas. El compuesto inhibió de fnorma notable el crecimeinto de estos dermatofitos, con un valor promedio de IC50 de 0.5μg/mL[6].

Se creó un modelo de infección por dermatofitos en cobayas, con lesiones de 2.5cm² de extensión. SE aplicó una solución al 10% de ME1111 sobre la zona afectada, durante siete días seguidos. Su efectividad clínica y antimicótica fue mucho mayor que la del grupo control con suero fisiológico y el grupo de referencia tratado con ciclopirox al 8%[7].

Experimentos celulares [1]:

Líneas celulares

Líneas celulares humanas K562, HepG2, U937, A431 (leukemia, liver tumor, lymphoma y skin origin, respectivamente)

Método de preparación

Las líneas celulares humanas se cultivaron en medio de crecimiento por debajo del 5% del CO₂ a 37°C. DEspués de 24 horas, se añadió ME1111 (a concentraciones que oscilaban entre 0.06 y 32μg/mL), seguido de una incubación de 72 horas.

Condiciones de reacción

0.06 a 32μg/mL; 72 hours.

Áreas de aplicación

ME1111 mostró una inhibición mínima de la proliferación celular human, con valores de IC₅₀ de 47, 37, 33 y 40μg/mL para las células K562, HepG2, U937 y A431, respectivamente. La selectividad para inhibir Trichophyton spp. (MIC₉₀=0.5μg/mL) sobre las células humanas fue 60 veces mayor, lo que indica una baja toxicidad de la célula huésped y un índice terapéutico favorable.

Conejillos de indias albinos machos (Harlan-Sprague-Dawley)

Método de preparación

Los conejillos de indias se infectaron aplicando una suspensión conidial que contenía 1x10⁷ T. mentagrophytes conidia en la piel abrasada de sus espaldas. El tratamiento con solución de ME1111 al 10% comenzó 72 horas después de la infección.

Forma de dosificación

Solución ME1111 al 10%; Aplicación tópica; Una vez al día durante siete días consecutivos.

Áreas de aplicación

El tratamiento con ME1111 condujo a una eficacia clínica y micológica significativa contra la dermatofitosis. La eficacia clínica de ME1111 fue superior a la del 8% de ciclopirox.
