RG 108 es un inhibidor potente de las DNA metiltransferasas, con un IC50 de 115nM[1]. Al actuar sobre estas enzimas, baja mucho la hipermetilación de lso promotores GSTP1, APC y RAR-β2 en células de cáncer de próstata, y esto afecta el avance del ciclo celular[2]. Se utiliza mucho para mejorar la eficiencia de la reprogramación de células madre pluripotentes inducidas (iPSCs) y favorecer la formación de blastocistos[3].
En cultivos celulares, RG 108 aplicado durante 12 horas frena la proliferación de células Eca-109 y TE-1, con IC50 de 70µM y 75µM, respectivamente[4]. Con 50μM durante 3 días, sube la expresión de NANOG, OCT4 y CD105 en células madre mesenquimales de médula ósea humana, y a la vez baja los niveles de transcripción de las DNMTs[5]. Al tratar con 100μM durante 5 días, corrige el fenotipo de senescencia en células madre del ligamento periodontal humano y fomenta la adipogénesis en estas células[6].
En estudios con animales, la inyección intraperitoneal de 20mg/kg/día durante 3 días atenúa mucho la lesión renal aguda causada por cisplatino en ratones, reduciendo la dilatación tubular y la deposición de glucógeno[7]. Una dosis única de 10mg/kg, inyectada 2 horas antes de la exposición al ruido, ayuda a mejorar la pérdida auditiva, reduce el daño a las células ciliadas y pérdida de sinapsisi auditivas en ratones[8].

Método de preparación | Las células Eca-109 se cultivaron en medio de águila modificada de Dulbecco (DMEM) suplementada con un 10% de suero bovino fetal (FBS), 100µg/ml de streptomycin y 100U/ml de penicillin en una incubadora humidificada a 37℃ con 5% de CO2. Las células se sembraron en placas de fondo plano de 96 pozos a una densidad de 2.5×103 células/pocillo y se trataron con diferentes concentraciones de RG 108 (5, 10, 25, 50, 75, 100, 150 y 200µM) durante 24 horas. Se añadieron 20μl de solución de MTT a una concentración de 5mg/ml a cada pocillo, y el cultivo se continuó durante 4 horas, y el probador midió la absorbancia a una longitud de onda de 490nm. |

Método de preparación | Los ratones machos C57BL/6J se mantuvieron en un entorno estéril estándar con acceso ad libitum a la dieta y el agua. treinta y seis ratones machos C57BL/6J (20-22g) se dividieron en cuatro grupos: grupo de control de solución salina, grupo de fármacos, grupo de incubacción de cisplatino y grupo de tratamiento RG 108. En el grupo de medicamentos, se inyectó RG 108 (20mg/kg) por vía intraperitoneal. En el grupo inducido por cisplatino, los ratones fueron inyectados por vía intraperitoneal con cisplatino (20mg/kg). En el grupo tratado con RG 108, se inyectó RG 108 a una concentración de 20mg/kg por vía intraperitoneal una vez al día durante tres días. Los ratones fueron anestesiados con isoflurano al 5%, y los riñones fueron extirpados para su análisis. |
