TAPI-1 es un inhibidor de la TACE (enzima convertidora del factor de necrosis tumral α). Y frena la liberación del dominio extracelular de la anfirregulina (AREG) al bloquear su actividad protelítica[1, 2]. También es inhibidor de metalloproteinase (MMP)[3]. De hecho, reduce el estrés oxidativo celular y la generación de factores inflamatorios[4].
En cultivos celulares, TAPI-1 (0-20µM) durante 24 horas bajó la viabilidad, migración e invasión de células TE-1 y Eca109 de forma dosis-dependiente, y potencia la apoptosis inducida por cisplatino[5]. Con 20µM en 24h, frenó la proliferación de células A549 y redujo la expresión de mRNA de EGFR y Notch1[6]. Con 5 y 10µM durante 6 y 24 horas, impidió la liberación de anfirregulina (AREG) de la membranan celular en células NCI-H292 estimuladas con diacetilo[7].
En animales, la inyección intraperitoneal de 10mg/kg de TAPI-1 en ratones con artritis por Staphylococcus aureus disminuyó mucho la hinchazón de las patas y los niveles de superóxido óxido nítrico y peróxido de hidrógeno en tejidos articulares[8].

Método de preparación | Las células TE-1 y Eca109 se expusieron a dosis variables de TAPI-1 (0, 1.25, 2.5, 5, 10 y 20μM) durante 24h, después de lo cual se determinó la viabilidad celular mediante el ensayo CCK-8. |

Método de preparación | TAPI-1 se diluyó en 5% de DMSO en solución salina estéril. La concentración final utilizada fue de 10mg/kg de peso corporal. La primera dosis de TAPI-1 se administró por vía intraperitoneal en cada ratón 24 horas después de la infección por S.aureus y se repitió el día, día 7, día 10 y día 13 después de la infección. Los ratones de control recibieron una sola dosis de TAPI-1 dos días antes del sacrificio. |
