أظهرت تجارب الفعالية في المختبر أنّ إعطاء calcitriol عن طريق الفم كل أسبوع يسمح بالتوصّل إلى أكبر تزكيرات calcitriol المصل وهو أعلى بكثير من 1 nM، وهذا التركيز يتمكن من تثبيط أكثر من 50% من تكاثر سرطان غدة البروستاتا. 10 nM من calcitriol قلل تعبير بروتين RAGE بشكل ملحوظ ورفع تركيزات sRAGE في خلايا عضلة القلب HL-1 مقارنة بخلايا التحكم[3]. أظهر Calcitriol التأثيرات المضادة للتكاثر ضد T47D وMCF-7 وMDA-MB-231 مع قيم IC50 بين 0.05-0.25 μM. بالإضافة إلى ذلك، يثبط calcitriol تكاثر خلايا ميلانوما مع IC₅₀ من 0.24 μM[5] . تم تثبيط نمو خلايا BT-474 بcalcitriol بشكل متدرج مع IC50 من 2.9 nM. إنّ 1 nM من Calcitriol عزّز بشكل منسّق التأثيرات المضادة للتكاثر AZD4547، مما يسمح بتقليل ضعفين الجرعة من AZD4547.
