Description of Genipin

Genipin est un inhibiteur spécifique de la protéine déouplante 2 (UCP2) extrait de Gardenia jasminoides Ellis[1]. Genipin a été largement utilisé pour la délivrance de médicaments en raison de sa biocompatibilité excellente, de sa biodégradabilité remarquable et de ses propriétés de réticulation stables[2]. Genipin a également été utilisé comme réactif pour la collecte d'empreintes digitales et comme agent de réticulation pour la préparation d'enzymes immobilisées [3][4].

Le traitement au Genipin (10mM, 2h) a entraîneé l'activation de l'IRS-1, de la PI3-K et de la signalisation en aval dans les cellules musculaires squelettiques de souris ( myoblastes C2C12) [5]. Genipin (200μM, 24h) induit l'apoptose dans les cellules de carcinome hépatique de rat FaO et dans les cellules humaines de carcinome hépatocellulaire Hep3B[6].

Genipin (25, 50, 100 et 200mg/kg; i.p.) a considérablement réduit la nécrose hémorragique, l'inflammation portale et la mortalité dans un modèle de souris de l'apoptose hépatique et de l'insuffisance hépatique [7]. Genipin (50mg/kg, 7 jours, p.o.) exerce des effets protecteur contre les lésions gastriques aiguës induites par l'éthanol chez les souris en inhibant l'activation du complexe inflammasome NLRP3[8].

Protocol of Genipin

Expériences cellulaires [1]:

Lignées cellulaires

Les cellules musculaires squelettiques de souris C2C12.

Méthode de préparation

Les cellules C2C12 ont été traitées avec 10 μM de Genipin penant 0.5h et 2h. Les lysats cellulaires ont été extraits avec un tampon de lyse (RIPA, 1×PBS, 1% Nonidet P-40, 5mM EDTA, 0.1% SDS, 1mM orthovanadate de sodium, 1% PMSF, cocktail d'inhibiteurs de protéases complet et inhibiteurs de phosphatases complets). Les lysats cellulaires complets ont été centrifugés à 12000 g à 4°C pendant 20min pour éliminer les matériaux insolubles. Pour l'immunoprécipitation, les surnageant ont été incubés avec un anticorps IRS-1 à 4°C pendant 16 h, puis avec des billes d'agarose-protéine A pendant 2 h. Les billes ont été lavées trois fois avec le tampon de lyse, bouillies avec le tampon d'échantillon de Laemmli pendant 5 min, puis chargées sur un gel SDS-PAGE;

Modèle de souris de l'apoptose hépatique et de l'insuffisance hépatique

Méthode de préparation

Les souris mâles ICR(20-22g) ont été privées de nourriture pendant la nuit masi avaient un accès libre à l'eau. Dans le groupe traité au Genipin, les souris ont reçu du Genipin (suspendu dans une solution saline de Tween 80 à 10%) à des doses de 25, 50, 100 et 200mg/kg par voie intrapéritonéale 1 heure avant le traitement à la d-galactosamine (GalN)/lipopolysaccharide (LPS), tandis que les autres groupes ont reçu un volume équivalent de solvant comme témoin. Tous les animaux ( sauf le groupe témoin normal) ont été injectés par voie intrapéritonéale avec GalN ( 700mg/kg) et LPS (10μg/kg Escherichia coli 026:B6) dissous dans une solution tamponée au phosphate.
