Description of QTZ

QTZ est un substrat bioluminescent utilisé pour l'imagerie in vivo. QTZ a une émission décalée vers le rouge et un faible bruit de fond. Comme analogue de la coelentérine, il porte une substitution 4-quinolyle en position C8. On peut l'utiliser dans les études sur l'imagerie in vivo et l'immunimagerie des tumeurs[1-2].

Protocol of QTZ

QTZ pour l'imagerie immuno-bioluminescence in vivo[1]

Ce protocole est adapté de données de recherche et fourni uniquement à titre de référence. Des ajustements pourront être nécessaires selon les exigences expérimentales spécifiques.

1.Préparation du réactif : Dissoudre QTZ dans un tampon d'injection in vivo pour préparer une solution stock à une concentration adaptée (peut être dissous jusqu'à ~2mM).

2.Préparation de l'enzyme appariée : Préparer la luciférase mutante QLuc, qui émet de la lumière ambrée et s'associe à QTZ. On peut y parvenir à l'aide d'un système de virus adéno-associé (AAV) porteur du gène d'expression de QLuc sous le contrôle d'un promoteur TBG spécifique du foie, ou par voie de construire, exprimer et purifier une protéine de fusion QLuc avec un fragement d'anticorps ciblant (par exemple, anti-Her2 scFv).

3.Modèle animal et marquage : Pour l'imagerie immuno-bioluminescente (ImmunoBLI), inoculer des cellules tumorales marquées à AkaLuc dans un modèle de rat xénogreffé. Injecter AAV exprimant QLuc (ou la protéine de fusion scFv-QLuc) par la veine caudale, ou administrer la protéine de fusion scFv-QLuc purifiée aux rats porteurs de tumeurs par injection intraveineuse.

4.Procédure d'imagerie : Une fois que l'expression de QLuc est établie dans le tissu cible (par exemple, le foie) ou que la protéine de fusion scFv-QLuc a circulé et s'est accumulée in vivo (par exemple, attendre 6h après l'injection de la protéine), administrer QTZ aux rats par injection intra-péritonéale ou intraveineuse (par exemple, 0,1µmol).

5.Acquisition du signal : A l'aide d'une caméra CCD refroidie ou d'un système d'imagerie bioluminescent équipé de filtre intrarouge lointain de 600-700nm pour réduire les interférences de fond), réaliser une imagerie in vivo sur les rats anesthésiés pour capturer la lumière ambrée émise par la réaction QLuc-QTZ (pic d'émission ~585nm).

Précautions :

(1)La paire luciférase-luciférine QLuc-QTZ émet une lumière décalée vers le rouge (pic ~585nm), avec plus de 52% de sa lumière totale émise ayant des longueurs supérieures à 600nm, améliore la pénétration dans les tissus profonds et le rapport signal/bruit pour l'imagerie in vivo.

(2)La luminosité intrinsèque in vitro de la paire QLuc-QTZ est d'environ 24% de celle de la paire NLuc-FRZ, mais plus de 60 fois plus brillante que la paire FLuc-D-luciférine. Dans un modèle de foie de rat, son signal luminescent durable (>30 minutes) surpasse d'autres mutants NLuc à petit domaine unique testé (teLuc, LumiLuc).

(3)La solubilité de QTZ dans l'eau est approximativement 62.5-125μM, et sa solubilité dans le tampon d'injection in vivo peut atteindre ~2mM.
