Méthode de préparation | Les cellules NCI-H929 ont été resuspendues dans un milieu RPMI 1640 supplémenté avec 10% de sérum de veau foetal, 2mM de glutamine et 1x de Pénicilline/Streptmycine, puis plaquées dans des plaques de culture cellulaire de 384 puits et traitéses avec des dilutions de Sec61-IN-1 dans DMSO/milieu. Les plaques ont été incubées à 37℃ avec de 5% CO2 pendant 48 heures. Après 48 heures, du Celltiter-Glo a été ajouté à chaque puits et le signal de luciférase a été quantifié pour déterminer la viabilité cellulaire. |
