Description of Shikonin

Shikonin est un produit naturel quinone - contenant. Shikonin est un inhibiteur puissant du canal chlorure TMEM16A qu'une valeur d'IC50 est 6,5µM[1]. Shikonin exerce un effet anti - inflammatoire par inhiber le facteur de nécrose tumorale -α (TNF-α) et empêche l'activation de la voie du facteur nucléaire -κB (NF-κB) via l'inhibition du protéasome. Shikonin inhibe l'activation d'inflammasome AIM2[2][3].

Shikonin (1µM) a induit l'apoptose capase - dépendante dans les cellules U937 après 6 heures, tout en augmentant la fragmentation de DNA, les radicaux oxydants intracellulaires (ROS), en baissant le potentiel du membrane mitochondriale, et les protéines pro - apoptotiques Noxa et tBid en exprimant[3]. Le traitement par 8µM Shikonin aux cellules A549 du cancer du poumon de cellules non - petites a significativement inhibé la prolifération cellulaire, a réduit l'expression de l'intégrine β1 et a inhibé la phosphorylation d'ERK1/2 à la tout au niveau de mRNA et de protéine[4]. Le traitement par Shikonin (10µM) aux cellules PC12 (phéochromocytome de rat) a protégé près de 70% cellules contre la toxicité, via les voies anti - apoptotiques directes et glutathion - dépendantes[5].

Le traitement local de Shikonin (1.0mg par oreille) a réduit la translocation iduite par 12 - O - tétracécanoylphorbol 13 - acétate (TPA) de la protéine kinase Cα ainsi que la phophorylation et l'activation d'ERK1/2, la transclocation nuléaire de NF-κB, et l'activité de NF-κB-DNA -liaison TPA - induite dans la peau de rat[6]. Dans le modèle de pancéatite aiguë induite par la pénicille des rats avec injury aiguë du poumon, Shikonin (50mg/kg) a réduit considérablement les scores histologiques pancréatiques ainsi que les activités de l'amylase et de la lipase sériques, et la production des cytokines TNF-α, IL-1β et IL-6 et l'activité de MPO ont été également significativement réduites[7]. Dans le modèle d'injury aiguë du poumon, Shikonin (50mg/kg) a considérablement réduit la production des cytokines pro - inflammatoires TNF-α et IL-1β ainsi que les concentrations de MPO et de monoxyde d'azote (NO) dans les rats avec une injury aiguë du poumon induite par le lipopolysaccharide (LPS). Le prétraitement par Shikonin aux rats a significativement inhibé l'activation LPS - induite de COX-2 et iNOS, ainsi que l'activité de liaison DNA NF-κB dans les tissus pulmonaires[8].

Méthode de préparation

Les cellules (MCF-7 et MCF-7/Adr) ont été injectées (s.c) dans un flanc de rats nus féminins âgés de 5 semaines. Le septième jour après l'inoculation, les rats ont été administrés par un véhicule de témoin ou Shikonin (2.5mg/kg/j, 5 jours, i.p) suspendue dans 20% itralipide dans un volume total de 0,2mL. Les rats ont été sacrifiés 2 jours après la dernière injection de Shikonin.
