Thidiazuron fait partie des dérivés d'urée disubstitués. Il possède une très forte activité cytokinine, on l'utilise beaucoup comme herbicide et régulateur de croissance végétale[1]. Thidiazuron agit sur plusieurs mécanismes de morphogenèse chez les plantes : on peut notamment citer l'apparition des tiges axillaires, la régénération de tiges adventices et l'embryogenèse somatique, sur un grand nombre d'espèces végétales[2].
Thidiazuron (0~50μM; 24h) réduit de manière dose-dépendante l'expression de uPA, uPAR, MMPs et VEGF dans les cellules MDA-MB-231[3]. Thidiazuron (0, 0.025, 0.05, 0.1 et 0.4mg/L; 4 semaines) augmente l'expression de la cytokinine oxydase[2]. Thidiazuron (0, 2.3, 4.5, 6.9 et 9μM; 10 semaines) stimule la formation d'embryons somatiques dans les explants de fraisier[4].

Méthode de préparation | Les cellules ont été cultivées sur les lames à quatre chambres, exposées à Thidiazuron (50μM) pendant 2h, fixées et perméabilisées. Les cellules ont été incubées avec un anticorps anti-p65, puis avec un anticorps secondaire marqué à FITC. La localisastion subcellulaire de la protéine p65 a été observée au microscope confocal à un grossissement de 40 fois. |
