Zosurabalpin est une nouvelle classe d'antibiotiques peptidiques macrocycliques liés, actifs contre divers isolats du complexe Acinetobacter baumannii (ABC), sa valeur deMIC90 est 0.06-0.5mg/L[1]. Zosurabalpin perturbe le transport du lipopolysaccharide (LPS) en inhibant le complexe protéique LptB2FGC, ce qui provoque l'accumulation de LPS au niveau toxique à l'intérieur de la cellule, entraînant finalement la mort bactérienne[2]. Zosurabalpin a été largement utilisé pour inhiber la prolifération de bactéries résistantes aux médicaments dans les modèles animaux[3].
In vivo, le traitement avec Zosurabalpin via injection sous-cutanée à la dose de 60mg/kg toutes les 6 heures pendant 24 heures a significativement réduit la charge bactérienne et amélioré la survie dans un modèle de rat de pneumonie induite par A. baumannii[4].

Méthode de préparation | Une neutropénie a été induite chez les rats CD-1 mâles par deux injections intra-péritonéales consécutives de cyclophosphamide monohydrochlorure (CPM) aux jours -4 et -1, respectivement, avant le début du traitement avec Zosurabalpin. Approximativement 106CFU de suspension bactérienne d'un isolat d' A. baumannii a été injectée dans chaque muscle fémoral pour induire une infection, et le traitement a débuté 2 heures après l'infection. Zosurabalpin (60mg/kg) a été administré par voie sous-cutanée toutes les 6 heures. La charge bactérienne fémorale a été déterminée 24 heures après le traitement. |
