Ani 9 es un inhibidor muy selectivo de ANO1, logra bloquear con gran potencia la actividad de este canal, con un IC₅₀ inferior a 3μM. Curiosamente, no altera las señales de calcio intracelular ni la función del canal de cloruro CFTR. Puede resultar una excelente herramienta farmacológica para investigar ANO1. Además, representa a un candidato prometedor para elaborar fármacos contra el cáncer, la hipertensión, el dolor, la diarrea y el asma[1].
En ensayos in vitro, al tratar células PC-3 con Ani 9 entre 3-30μM durante 24-72h, se frena su crecimeinto, según la dosis y el tiempo de exposición[2]. Por otro lado, al emplear hasta 30μM de este compuesto durante 20 minutos, no modifica el aumento de calcio citosólico provocado por el ATP en células FRT[1]. En las células LN215, después de 30 minutos de tratamiento, las concentraciones de 1μM, 3μM y 10μM de Ani 9 redujeron la actividad de VRAC en un 13.5±1.1%, 21.4±1.0% y 40.7±1.5%, respectivamente[1]. Con 3μM de Ani9 y 30 minutos de incubación, no se observan cambios en las contracciones del músculo liso uretral de ratón macho generadas por la estimulación de campo eléctrico (EFS)[3]. Asimismo, 10μM de Ani 9 aplicados durante media hora no consiguen inhibir las contracturas de bronquios murinos desencadenadas por carbacol[4].
