Description of Apyrase

Apyrase es una nucleoside triphosphate diphosphohydrolase (NTPDase) que elimina catalíticamente el fosfato terminal de los NTPs y NDPs [1]. La actividad de la Apyrase vegetal se ha estimulado de forma notable por la calmodulina activada por Ca2+[1]. Una unidad de Apyrase se define como la cantidad de enzima que libera 1.0μmol de fosfato inorgánico por minuto a partir de ATP o ADP, a pH 6.5 y 30°C.

Apyrase (20U/mL; 6h) redujo los niveles de RecA en Escherichia coli O157:H7; A concentración de 10 y 20U/mL (6h) disminuyó la cantidad de DNA del fago Stx2 en sobrenadantes de esta bacteria, de manera dependiente de la dosis. A 10U/mL (6h), redujo la libreación de la toxina Shiga 2 por E. coli O157:H7 in vitro [2].

Apyrase (15U; 0, 2d; i.p.) dimó la aparición de síntomas en ratones BALB/c infectados con E. coli O157: H7. Asimismo, redujo la apoptosis de células intestinales y los niveles de ATP en heces de estos animales[2]. Apyrase (2U; i.p.) disminuyó de forma el tamaño tumoral en ratasWistar con glioma implantado [3]. Apyrase (4U/g; i.v.) redujo el número de neutrófilos y macrófagos en el cerebro isquémico de ratones Cd39 knock-out, así como el tamaño del infarto cerebral[4]. Apyrase (0.2U; top) disminuyó los niveles de ATP en la piel de ratones C57BL/6 después de una quemadura. Administrada durante 2 días, inhibió la reproducción bacteriana en las zonas quemadas[5].

Método de preparación

E. coli O157:H7 se incubó con o sin apyrase durante 6h y se analizaron lisados bacterianos para detectar el nivel de RecA.

Método de preparación

La piel local de los ratones se sumergió en agua destilada a 60°C para producir una quemadura de escaldadura de espesor parcial y luego se infectó con Acinetabacter baumannii resistente a los antibióticos inmediatamente. Un tratamiento diario de 200 μL (1U/mL) de apyrase o vehículo solo se aplicó a la herida de quemadura. Después de 2 días, se cosechó la piel para cuantificar las CFUs.
