Description of Apyrase

Apyrase는 NTP와 NDP의 말단 인산을 촉매적으로 제거하는 뉴클레오사이드 삼인산 디인산수소분해효소(NTPDase)의 일종이다. 식물 Apyrase의 활성은 Ca²⁺-활성화 칼모듈린에 의해 현저하게 증진된다 [1]. 1단위는 pH 6.5, 30 °C 조건에서 ATP 또는 ADP로부터 1.0 μmol의 무기 인산을 분당 방출할 수 있는 Apyrase의 양으로 정의된다 [1].

Apyrase(20 U/mL, 6시간)는 O157:H7 대장균에서 RecA 수준을 감소시켰다; Apyrase(10, 20 U/mL, 6시간)는 O157:H7 대장균 상등액 내 Stx2 파지 DNA 양을 용량 의존적으로 감소시켰다; Apyrase(10 U/mL, 6시간)는 체외 실험에서 O157:H7 대장균의 Shiga 독소 2 방출을 감소시켰다 [2].

Apyrase(15 U, 0, 2일차, i.p.)는 O157:H7 대장균에 감염된 BALB/c 쥐의 증상 발현을 지연시켰다; Apyrase(15 U, i.p.)는 감염된 BALB/c 쥐의 장 세포 자멸사를 감소시켰다; Apyrase(15 U, i.p.)는 감염된 BALB/c 쥐의 분변 내 ATP를 낮췄다 [2].Apyrase(2 U, i.p.)는 이식된 교모세포종이 있는 Wistar 쥐에서 종양 크기를 현저하게 줄였다 [3].Apyrase(4 U/g, i.p.)는 Cd39 녹아웃 쥐의 허혈성 뇌 내 호중구와 대식세포 수를 감소시키고 뇌경색 크기를 줄였다 [4].Apyrase(0.2 U, 국소 도포)는 화상 후 C57BL/6 쥐 피부의 ATP를 감소시켰다; Apyrase(0.2 U, 2일간, 국소 도포)는 C57BL/6 쥐 화상 부위의 세균 증식을 억제하였다 [5].

제조 방법

E. coli O157:H7을 apyrase 유무에 따라 6시간 배양한 후 세균 용해물에서 RecA 수준을 측정하였습니다.

제조 방법

쥐 국부 피부를 60°C 증류수에 담가 부분 두께 화상을 유발한 후 즉시 항생제 내성 Acinetobacter baumannii로 감염시켰습니다. 화상 상처에 매일 200 μL(1 U/mL)의 apyrase 또는 용매만을 국소 도포하였고 2일 후 피부를 채취해서 CFU를 정량하였습니다.
