Description of Apyrase

Apyrase is a kind of nucleoside triphosphate diphosphohydrolase (NTPDase) that catalytically removes the terminal phosphate from NTPs and NDPs [1]. The activity of plant Apyrase is significantly stimulated by Ca2+-activated calmodulin [1]. One unit is defined as the amount of Apyrase that can release 1.0μmol of inorganic phosphate from ATP or ADP per minute at pH 6.5 and 30°C.

Apyrase (20U/mL; 6h) decreased the levels of RecA in Escherichia coli O157:H7; Apyrase (10, 20U/mL; 6h) decreased the amount of Stx2 phage DNA in E. coli O157:H7 supernatants in a dose-dependent manner; Apyrase (10U/mL; 6h) decreased Shiga toxin 2 release from E. coli O157:H7 in vitro [2].

Apyrase (15U; 0, 2d; i.p.) delayed the development of symptoms in BALB/c mice infected with E. coli O157: H7; Apyrase (15U; i.p.) decreased the apoptosis of intestinal cells of the BALB/c mice infected with E. coli O157:H7; Apyrase (15U; i.p.) decreased the ATP in feces from E. coli O157:H7-infected BALB/c mice [2]. Apyrase (2U; i.p.) significantly reduced the size of tumor in the Wistar rats with implanted glioma [3]. Apyrase (4U/g; i.v.) decreased the number of neutrophils and macrophages in the ischemic brain of Cd39-null mice and diminished the size of cerebral infarct[4]. Apyrase (0.2U; top) reduced ATP on skin of C57BL/6 mice after burn; Apyrase (0.2U; 2d; top) inhibited bacterial reproduction in the burns of C57BL/6 mice [5].

Preparation Method

E. coli O157:H7 was incubated with or without apyrase for 6h and bacterial lysates were assayed to detect the level of RecA.

Preparation Method

The local skin of mice was immersed in 60°C distilled water to produce a partial-thickness scald burn and then were infected with antibiotic-resistant Acinetabacter baumannii immediately. A daily treatment of 200 μL (1U/mL) of either apyrase or vehicle alone was applied topically to the burn wound. After 2d, skin was harvested to quantify CFUs.

Applications

Apyrase inhibited bacterial reproduction in the burns of C57BL/6 mice.
