CA77.1 es un derivado de molécula pequeña oralmente activo de 7-chloro-3-(p-tolyl)-2H-benzo[b][1,4]oxazine (AR7) y puede actuar como activador de la autofagia mediada por chaperona (CMA)[1]. CA77.1 ejerce efectos neuroprotectores y se está desarrollando para el tratamiento de varias enfermedades crónicas del CNS, incluida la enfermedad de Alzheimer, la enfermedad de Parkinson y la degeneración retiniana[2].
In vitro, CA77.1 (12μM) pretrató las células BV2 microgliales murinas durante 12h seguidas de tratamiento con LPS. CA77.1 disminuyó la expresión de iNOS y COX-2 inducida por LPS, redujo los factores proinflamatorios NO e IL-6 y redujo la expresión de mRNA de iNOS, COX-2, IL-6 e IL-1β sin afectar la viabilidad celular[3]. El tratamiento de CA77.1 (20µM) en células de neuroblasto Neuro-2a (N2a) de ratón durante 24h promovió la degradación de GAPDH (un sustrato canónico de CMA) y aumentó la eliminación de subunidades de Na+ /K+ -ATPase β1, acompañado de una disminución de la actividad de Na+ /K+ -ATPase y un aumento de la concentración de Na+ intracelular, sin cambios en los niveles de mRNA[4]. El tratamiento CA77.1 (0-30μM) en células renales porcinas PK-15 que expresan la proteína no estructural 3D del virus de la enfermedad de la fiebre aftosa (FMDV) durante 24h disminuyó efectivamente los niveles de polimerasa 3D de FMDV y suprime la replicación de FMDV a través de la activación de la vía de la CMA[5].
In vivo, CA77.1 (10mg/kg/día) se inyectó intraperitonealmente en modelos de ratones de hemorragia intracerebral (ICH) durante seis días consecutivos. La administración de CA77.1 revirtió el aumento inducido por la ICH de los astrocitos reactivos A1, el daño de la mielina, la muerte neuronal y los trastornos neuroconductuales [6]. CA77.1 (10mg/kg) se administró por vía intraperitoneal en ratones expuestos a la toxina mitocondrial BDE-47 con deterioro cognitivo cada tres días durante 4 semanas. CA77.1 mitigó el deterioro cognitivo inducido por BDE-47 en ratones al suprimir la ferroptosis, prevenir la pérdida sináptica en los hipocampos de ratones y rescatar la atrofia estructural neuronal del hipocampo[7].

Método de preparación | Los ratones se dividieron aleatoriamente en los siguientes grupos: grupo de control, grupo 2,2',4,4'-éter tetrabromodifenilo (BDE-47), grupo BDE-47 + CA77.1. Los ratones del grupo BDE-47 recibieron por vía oral de 20 mg/kg/día BDE-47 y también se inyectaron por vía intraperitoneal con solución salina normal que contenía un 2 % de DMSO durante 8 semanas. Los ratones del grupo BDE-47 + CA77.1 recibieron una inyección intraperitoneal de 10 mg/kg de CA77.1 cada tres días durante las últimas 4 semanas antes de la administración de 20 mg/kg/día de BDE-47. |
