Guadecitabine sodium es un novedoso dinucleótido hipometilante de decitabina y desoxiguanosina que es resistente a la degradación por citidina desaminasa. Guadecitabine Sodium es la forma fácilmente disoluble de Guadecitabine[1].
Guadecitabine (0.1, 0.3, 1, 5µM, 48h) aumentó la sensibilidad al cisplatino tanto en las células parentales A2780 como en las resistentes. Aunque entre otras líneas celulares de cáncer de ovario, las células parentales A2780 resistentes al cisplatino se consideran "sensibles" al cisplatino, tienen una IC50 relativamente alta para el fármaco[2].
El pretratamiento con Guadecitabine (50nM-2µM, 24h) interactuó sinérgicamente con ASTX660 para inducir la muerte celular en cinco líneas celulares de LMA (MOLM-13, ML-2, MV4-11, PLB-985, KG-1) con diversos antecedentes genéticos y que representan diferentes subtipos de LMA[3].
Ratones inmunodeficientes portadores de tumores fueron expuestos subcutáneamente a Guadecitabine a dosis de 3, 6.1, o 10 mg/kg, diariamente durante 5 días, con tumores recolectados en el día 7. La mayoría de los ratones tratados en el esquema de 5 días con 10 mg/kg/día de Guadecitabine murieron; todos los ratones tratados con 6,1 mg/kg/día de Guadecitabine desarrollaron toxicidad gastrointestinal. Se observó toxicidad mínima en los ratones tratados con 3 mg/kg/día. El tratamiento con Guadecitabine causó hipometilación de LINE-1 y NY-ESO-1 en todas las dosis[4].

La combinación de Guadecitabina y células T específicas para NY-ESO-1 mostró un retraso en el crecimiento tumoral en comparación con los ratones tratados solo con Guadecitabina o células T CD8+ específicas para NY-ESO-1. Estos datos sugieren que el tratamiento con Guadecitabina potencia las respuestas antitumorales específicas para NY-ESO-1 in vivo. |
