R243 es un inhibidor del receptor CCR8 que frena las vías de señalización activadas por CCL1, su ligando endógeno[1-2]. Se utiliza sobre todo en estudios de enfermedades inmunes e inflamatorias[3].
En cultivos celulares, R243 (10µM) durante 24 horas redujo mucho la migración quimiotáctica por CCL1 (10ng/mL) en macrófagos RAW264.7 y macrófagos derivados de médula ósea de ratón. Aunque su efecto inhibidor en monocitos humanos THP-1 no fue muy notable[4]. En macrófagos peritonales de ratón estimulados con LPS (100ng/mL), R243 (0–10µM) durante 24 horas bajó la secreción de TNF-α, IL-6 e IL-10. También redujo la fosofrilación de las vías JNK y NF-κB[5].
En animales, en un modelo de fibrosis peritoneal, R243 a 1.0mg/kg inyectado tres veces por semana durante 28 días alivió mucho la inflamación y fibrosis peritoneales inducidas por gluconato de clorhexidina. Lo notamos en menor grosor peritoneal, menos deposición de colágeno y menos infiltracción de macrófagos[6]. En pruebas de hiperalfgesia térmica, R243 entre 0.3–1mg/kg inyectado 30 minutos antes de CCL1 frenó de forma dosis-dependiente la analgesia sistémica provocada por CCL1 (10µg/kg)[7].

Método de preparación | Las células se mantuvieron en un medio RPMI 1640 suplementado con 10% de suero bovino fetal (FBS), 2mM de L-glutamine y 1% penicillin/streptomycin a 37°C, 5% de CO₂. Para los ensayos de migración de transwell, las células se activaron con R243 (10μM), LPS (100ng/mL) e IFN-γ (10ng/mL) y se sembraron en insertos de transwell. Se añadió CCL1(10ng/mL) en los desembastes de transwell. Para el análisis de la expresión génica, los macrófagos RAW264.7 se activaron con R243 (10μM), CCL1(10ng/mL), LPS (100ng/mL) e IFN-γ (10ng/mL). |

R243 también suprimió la expresión génica de los marcadores proinflamatorios (iNOS e IL-6) y CCR8 en macrófagos RAW264.7 activados por LPS/IFN-γ. |

Método de preparación | La fibrosis peritoneal (PF) se indujo en ratones a través de una inyección intraperitoneal diaria de solución de gluconato de chlorhexidine gluconate (CG) de 0.2mL (compuesta por 0.1% de CG y 15% de etanol) durante 4 semanas. Ratones fueron tratados con R243 (1.0mg/kg) a través de inyección intraperitoneal tres veces por semana, comenzando desde el día 0 y continuando hasta el día 28. El días 28, se sacrificaron ratones y se cosecharon tejidos petitoneales para su análisis. |
