SPD304 actúa como inhibidor selectivo del factor de necrosis tumoral alfa, o TNF-α. Su valor de IC50 es de 22μM[1]. Esta molécula hace que los trímeros de TNF se separen. De esta manera, impide que la citocina se una a sus receptores celulares[2]. Se suele emplear para bloquear la ruta TNF-α/NF-κB/CUL4B y detener el ciclo celular en la fase S [3].
Al tratar células SPD304 con 30µM durante 24 horas, se observa una marcada muerte celular[4]. Una concentración de 25µM aplicada durante el mismo lapso reduce la actividad del TNFα y disminuye la viabilidad de las células L929[5]. Las células GH3 expuestas a 8µM del compuesto durante 48 horas dejan de elevar las proteínas de la ruta MAPK por acción del TNFα. También se frena su capacidad de multiplicar, moverse e invadir teidos[6]. Si se preincuban fibroblastos dérmicos humanos con 1µM de SPD304 durante 24 horas, bajan los niveles de IL-8 e IL-6 en estas células (HDFs)[7].
En ratones con insuficiencia cardíaca, se administró SPD304 mediante inyección intraventricular cerebral. Se utilizan 5µg del compuesto disuelto en 10µl de líquido cefalorraquídeo artificial con un 5% de DMSO, y la aplicación duró cinco minutos. El tratamiento neutralizó el aumento de la presión arterial, la aceleración del ritmo cardíaco y el aumento de la actividad simpática renal provocados por la enzima convertidora del TNF-α (TACE)[8].

Método de preparación | Las células HEK293T se chaparon en placas de 96 pozos a 1×104 células por pozo y se cultivaron en 100μl de DMEM a 37°C durante 24 horas. Luego, a cada pozo se añadieron 50μl de SPD304 en varias concentraciones (1, 10, 20, 40, 60, 80 y 100µM). En los grupos de control, se utilizaron 50μl de DMEM. Los pozos que contenían solo medios sin células y aditivos se utilizaron como espacios en blanco. La viabilidad celular se midió después de 24 horas. |

Método de preparación | Las ratas macho de Sprague-Dawley rats (225-275g) fueron alojadas en una instalación de cuidado de animales con temperatura (23±2°C) y luz controlada (ciclo de 12:12 horas luz-oscuridad). La insuficiencia cardíaca (HF) y las ratas simuladas (Sham) fueron producidas por ligadura de la arteria coronaria o cirugía simulada. Las ratas Sham (n=6) y HF (n=6) fueron tratadas con inyección intratracerebroventricular de TACE (300ng en 3µl de líquido cefalorraquídeo artificial durante 1 minuto) immediatamente después del pretratamiento intratracerebroventricular con el SPD304 (5µg en 10µl 5% DMSO en líquido cefalorraquídeo artificial durante 35 minutos). Se analizaron la presión arterial media (MAP), la frecuencia cardíaca (HR) y la actividad del nervio simpático renal (RSNA). |
