Boc-Phe-Ser-Arg-AMC est un substrat fluorescent qui, une fois clivé par des protéases, libère AMC hautement fluorescent (longueurs d'onde d'excitation/émission : 355 nm/460 nm). Boc-Phe-Ser-Arg-AMC est caractérisé par sa dégradation préférentielle par le facteur de coagulation Xla et est largement utilisé pour détecter l'activité d'enzymes telles que la typsine, la tryptase et la kallicréine[1,2,3]. Boc-Phe-Ser-Arg-AMC est également très sensible aux protéases de la matrice mitochondriale de la levure et à la trypsine des mastocytes de rat. Lorsque la liaison peptidique Arg-AMC est clivée par les protéases, AMC est libéré, et l'augmentation de l'intensité de fluorescence est proportionnelle à l'activité enzymatique[4,5,6].

Utilisation de Boc-Phe-Ser-Arg-AMC pour mesurer l'activité trypsine-type dans les échantillons de stratum corneum[1]:
(1)Mélanger 0,5mg d'échantillons lyophilisés de stratum corneum avec 500µL de tampon d'essai (50mM Tris, 1M NaCl, 10mM EDTA, pH 8.5), incuber à 37°C pendant une heure sur un agitateur, puis centrifuger à 13000×g pendant 10min à 4℃.
(2)Récupérer le surnageant pour l'essai d'activité protéase; utiliser Boc-Phe-Ser-Arg-AMC comme substrat pour mesurer l'activité de la protéase trypsine-type.
(3)Mélanger 100µL d'échantillon de surnageant avec 100µl de tampon d'essai contenant 0,75mM de Boc-Phe-Ser-Arg-AMC, incuber à 37°C pendant 5h, et mesurer l'activité toutes les heures.
(4)Détecter AMC libéré à l'aide d'un détecteur de microplaques multifonctionnel aux longueurs d'onde Ex/Em de 355nm/460nm, et étalonner à l'aide d'étalons d'AMC.
Utilisation de Boc-Phe-Ser-Arg-AMC pour mesurer l'activité de la peptidase liée à la kallicréine 6 (KLK6)[2]:
(1)Ajouter 5µL d'inhibiteur (utiliser des dilutions 1:2 ou 1:3 pour obtenir différentes concentrations finales d'essai) ou un témoin DMSO (concentration finale d'essai 2%) dans chaque puits d'une microplaque noire de 384 puits.
(2)Ajouter ensuite 5µL de KLK6 (concentration finale d'essai 5nM) dissoute dans un tampon de réaction (50mM Tris, 150mM NaCl, 1mM EDTA, 0.05% Tween-20, pH 7.5), et incuber à température ambiante pendant 30 minutes.
(3)Juste avant de commencer la mesure cinétique de l'intensité de fluorescence (Ex, 350 ± 15nm; Em, 440 ± 20nm; toutes les 60 secondes pendant 30 minutes), ajouter 10µL du substrat Boc-Phe-Ser-Arg-AMC (concentration finale d'essai 20µM) dissous dans le tampon de réaction dans chaque puits.
(4)Calculer la pente des signaux mesurés à l'aide de GraphPad Prism, la normaliser par rapport au témoin DMSO (activité à 100%), puis l'utiliser pour générer des courbes dose-réponse.
