L-Albizziine est un inhibiteur de 4-méthylèneglutaminase, sa valeur d'IC50 est 20mM [1]. L-Albizziine, un inhibiteur de la glutamate synthase, a fortement inhibé la conversion du 2-cétoglutarate dans les feuilles de maïs[2]. L-Albizziine inhibe l'activité de l'asparagine synthétase et a été largement utilisé pour réguler l'activation des lymphocytes T dans différents modèles cellulaires[3].
Le traitement in vitro avec L-Albizziine à 2mM pendant 72 heures a significativement réduit le niveau d'aspartate et augmenté l'expression de la protéine p53 dans les cellules HuCCT1[4]. Le traitement avec 4mM de L-Albizziine pendant 48 heures a inhibé la prolifération des cellules HCT116 p53+/+ et p53−/− et réduit le nombre de cellules[5]. Le traitement avec 10mM de L-Albizziine pendant 24h a perturbé la réplication du virus de la variole dans les fibroblastes primaires de la peau humaine sans affecter la viabilité cellulaire[6].

Méthode de préparation | Les cellules HuCCT1 transfectées avec FGFR2-AHCYL1 (FA Cho-HDO) (200nM) ont été cultivées dans du milieu RPMI-1640 avec ou sans L-Albizziine (2mM) ou Asn (0.3mM) pendant 72h, le milieu était complété par 10% de sérum de veau foetal (SVF) et 1% de pénicilline/streptomycine, et les cultures ont été incubées dans une atmosphère humidifiée avec 5% CO2. Les protéines cellulaires ont été extraites et soumises à une analyse par Western Blot en utilisant un tampon de lyse RIPA contenant des inhibiteurs de protéases/phosphatases. |
