proTAME est un inhibiteur d' APC/C (Anaphase Promoting Complex/Cyclosome) perméable à la cellule[1]. proTAME est converti en TAME (Tosyl-L-Arginine Methyl Ester) par des estérases intracellulaires, et TAME inhibe spécifiquement l'activité d' APC/C, ainsi donc, induit un arrêt du cycle cellulaire en métaphase et entraîne ensuite la mort cellulaire[2]. proTAME est communément utilisé dans la recherche de la régulation du cycle cellulaire et les recherches liées au cancer[3][4].
Le traitement in vitro avec proTAME (5, 10, 20, 50 or 100μM; 3h) montre un arrêt en métaphase dose-dépendant dans les ovocytes mammaliens et les embryons de clivage précoce, et l'arrêt en métaphase induit par ce médicament ne nécessite par l'activité du point de contrôle de l'assemblage du fuseau (SAC)[5]. Le traitement avec proTAME(5, 10, 20, 50 or 100μM; 6h) aux cellules OVCAR-3 inhibe la croissance cellulaire, sa valeur d'IC50 est 12,5µM[6]. Le traitement avec proTAME (3, 6 et 12µM) aux lignées cellulaires MCL et DLBCL pendant 24h induit une métaphase prolongée, entraîne l'accumulation du substrat APC/C-Cdc20, la cycline B1, l'inactivation/dégradation de Bcl-2 et Bcl-xL et une apoptose dépendante de la caspase[7].

Méthode de préparation | Toutes les lignées cellulaires de MCL (Jeko-1, Mino and Rec-1) et ABC–DLBCL (U2932 et RI-1) ont été maintenues dans le milieu RPMI-1640 supplémenté avec 10% de sérum de veau foetal (FCS) et 2mM de glutamine. Les cellules ont été cultivées à 37°C dans une atmosphère humidifiée à 5% de CO2 et traitées pendant 24 heures avec proTAME (3, 6 et 12µM). L'effet sur la viabilité a été déterminé à l'aide d'un test CellTiter-Glo et les effets sur les substrats des co-activateurs de APC/C Cdc20 et Cdh1 ont été étudiés par Western Blot. |
