W146 est un antagoniste sélectif des récepteurs de la sphingosine 1-phosphate de type 1 (S1PR1), sa valeur d'EC50 est 398nM[1].W146 est largement utilisé pour étudier les processus biologiques médiés par S1P/S1PR1.
In vitro, W146 (10μM) a été utilisé pendant 30 minutes dans les cellules HEK293 et CHO-K1 exprimant de manière stable le récepteur S1P. W146 a complètement aboli l'internalisation, la phosphorylation et l'ubiquitinylation de S1PR1 induites par l'agoniste S1PR1 hauetement sélectif hydrosoluble CYM-5442 ou par S1P, ainsi que l'activation de p42/p44 MAPK[2]. La microglie a été pré-traitée avec W146 (1µM) pendant 30 minutes avant le traitement avec siponimod, puis les cellules ont été stimulées avec 1µg/mL de LPS seul (pendant 24h) ou 1µg/mL de LPS (pendant 3.5h) plus 10µM de l'activateur de l'inflammasome nigericine (pour les 30 dernières minutes). Le pré-traitement avec W146 et siponimod a significativement supprimé la production d'interleukine-1β dans la microglie activée stimulée par le lipopolysaccharide plus la nigericine[3].W146 (10μM) a été appliqué à des cellules endothéliales de l'artère coronaire humaine pendant 10 minutes, puis les cellules ont été exposées à S1P ou à des milieux contenant des protéines plasmatiques pendant 2 heures. W146 a aboli le rôle protecteur de S1P et des protéines plasmatiques sur la glycocalyx[4].
In vivo, W146 (10mg/kg) a été administré par voie intra-péritonéale à des rats Swiss non-jeûnés et a induit une lymphophénie chez les rats, ainsi qu'une augmentation parallèle des lymphocytes CD4+ et CD8+ dans les ganglions lymphatiques[5].W146 (0.1mg/kg) a été administré à des rats C57BL/6 via injection intra-péritonéale quotidiennement pendant 3 jours avant que les animaux ne soient soumis à une ischémie rénale de 20 minutes suivie d'une reperfusion de 24 heures. Les rats traités avec W146 ont présenté des lésions rénales et hépatiques significativement exacerbées[6]. W146 (5mg/kg) injecté par voie intra-péritonéale 1 heure avant l'administration d' AMD3100 a significativement amélioré la mobilisation des cellules souches et progénitrices hématopoïétiques Kit+/Sca-1+/Lin− (KSL) chez les rats C57BL/6 d'environ 8 fois[7].
