Fluorescent probes that bind with calcium ions and exhibit a light channel reaction allow researchers to observe changes in intracellular free calcium ion levels, which can be detected through English light microscopy, flow cytometry, fluorescence spectrophotometer, and fluorescence enzyme assay plates.

The new generation of calcium ion fluorescent probes: Cal-520, Ca-590, and Cal-630 are the strongest and most uniform fluorescent probes for calcium mobility measurement. Compared with traditional calcium ion probes (such as Fluo-3AM, Fluo-4 AM, and Rhod-2 AM), they have significantly improved signal-to-noise ratio and intracellular retention Ca-520 AM, Cal-590 AMCa-630 AM have fine permeability and pre enter fine. At first glance, the photophilic group AM is cleaved by enzymes, producing negatively charged Anglo dyes that remain. Once combined with calcium ions, their fluorescence is significantly enhanced. When cells are stimulated by agonists, receptors emit signals of intracellular calcium release, thereby increasing the fluorescence of Ca1-520, Cal-590, and Ca-630 inside the cells. The high sensitivity and>100 fold fluorescence increase of Ca-520 AM and Cal-590 AMCa1-630 AM make them ideal probes for intracellular calcium ion determination. Their high signal-to-noise ratio and heavy internal retention make them excellent tools for evaluating GPCR and calcium channel targets, as well as screening corresponding agonists and antagonists

Ca-520, Ca-590, and Ca1-630 are mainly located in the cytoplasm, unlike Rhod-2, which is mainly located in mitochondria. The long excitation/emission wavelength of Ca-590Ca-630 makes it a perfect calcium ion probe that can be labeled with green fluorescent protein (GFP) for detailed and multicolor detection. In addition, Ca-520, Ca-590, and Cal-630 are also compatible with the vast majority of existing fluorescence instruments.
