(Gly¹⁴)-Humanin (human)

(Gly14)-Humanin (human)은 세포 내 단백질이자 분비 단백질이다.

이는 미토콘드리아 유전체에서 인코딩된 첫 번째 새로 발견된peptide로, 많은 세포 유형에서 다량의 세포 독성 자극에 대한 강력한 보호 작용을 발휘한다^[2]. HN 肽 사슬의 14번째 위치에 있는 세린(Ser14)을 glycine (Gly)으로 교체하면 (Gly14)-Humanin (human)이 형성되며, Humanin에 비해 그 세포 보호 활동이 1000배 향상된다^[3].

(Gly14)-Humanin (human)은 RAW 264.7 거대세포에서 Dil-ox-LDL 축적을 감소시켰다. (Gly14)-Humanin (human)은 ox-LDL 유발의 거대세포 유래의 버블 세포 형성을 억제할 수 있었으며, 이는 섬유질 흡수 감소와 거대세포 세포의 콜레스테롤 외부 유출 증가로 인해 발생했다^[7]. (Gly14)-Humanin (human)전처리는 miR-155를 다운 조정하여 HG 유발의 혈관 내피세포의 세포 사망을 억제하고, 이는 세포 유래의 EMP를 통해 인접한 혈관 내피세포로 전달되었다. EMP는 miRNAs의 특이성 전달을 위한 잠재적 도구가 될 수 있으며, (Gly14)-Humanin (human)의 항당뇨병 혈관 합병증 치료에서 더 강한 치료 효과에 기여한다^[1]. (Gly14)-Humanin (human)은 HUVECs에서 Ox-LDL에 의한 림프질 기능 손상을 복구하기 위해 카테프시딘 D의 활성 및 단백질 수준을 회복하고, 더 나아가 HUVECs에서 Ox-LDL-induced의 자가포식 손상을 복구할 수 있었다^[4].

(Gly14)-Humanin (human)의 사전 처리는 공간 기억을 보호할 뿐만 아니라 Aβ 유발의 흐름으로부터 STAT3을 구해냈으며; (Gly14)-Humanin (human)의 신경 보호 효과는 제니스타인에 의해 효과적으로 중화되었으며, 제니스타인은 특이적tyrosine kinase genistein inhibitors이다. Aβ의 31-35 시퀀스는 분자에서 행동에 이르기까지 Aβ의 신경 독성을 가진 가장 짧은 활성 센터이다; (Gly14)-Humanin (human)은 Aβ 유발의 공간 학습 및 기억 손상을 대쥐로부터 보호한다; 타로신 효소의 활성화 및 그 후 STAT3과 caspase3에 대한 유익한 조절이 관여할 수 있다^[5]. TBI전에 뇌실내(intracerebroventricularly, i.c.v.)로 (Gly14)-Humanin (human)을 투여한 쥐는 손상된 피질과 해마에서 플라즈마막 펜메이빌리티 셀을 감소시켰으며, 뇌 손상 부피를 감소시키고, 운동 성능을 개선하고 TBI 후의 Morris 물 미궁 테스트에서 성능을 개선했다. TBI 후 1시간과 2시간에 복강내(intraperitoneally, i.p.) (Gly14)-Humanin (human)을 투여하였을 때에도 부상 부피가 감소되었으며, 운동과 Morris 물 미궁 테스트 결함에서 약간의 개선이 관찰되었다. (Gly14)-Humanin (human) 치료는 쥐의 TBI 후의 형태학적 및 기능적 결과를 개선했으며, (Gly14)-Humanin (human)의 TBI에 대한 보호 효과는 down-regulated apoptosis 및 autophagy와 관련이 있을 수 있다^[6].
