Description of Chitoheptaose

Chitoheptaose is a chitosan oligosaccharide monomer with a degree of polymerization of 7. Chitoheptaose exhibits anti-inflammatory and antioxidant activities[1].

In vitro, Chitoheptaose (0-400μM; 24h) exhibited no significant cytotoxicity in RAW 264.7 macrophages[1]. Chitoheptaose (200μM; 30-48h) significantly inhibited LPS-induced NO production, reduced IL-6 and TNF-α secretion, and suppressed the overexpression of p-IκBα in RAW 264.7 macrophages[1]. Chitoheptaose (200-800μg/mL; 24h) showed no significant cytotoxicity in HaCaT human keratinocytes, but exhibited certain toxicity at 1000μg/mL[2].

Protocol of Chitoheptaose

Cell experiment [1]:

Cell lines

RAW 264.7 macrophages

Preparation Method

RAW 264.7 macrophages were plated in 96-well plates at a density of 5×105cells per well and incubated for 24h. Then, 100μL of Chitoheptaose at varying concentrations (0-400μM) was added to the cells and incubated for an additional 24h.

Reaction Conditions

0-400μM; 24h

Applications

Chitoheptaose did not inhibit cell viability at concentrations up to 400μM.

Calculation results:

Working concentration: mg/ml;

Method for preparing DMSO master liquid: mg drug pre-dissolved in μL DMSO ( Master liquid concentration mg/mL, Please contact us first if the concentration exceeds the DMSO solubility of the batch of drug. )

Method for preparing in vivo formulation: Take μL DMSO master liquid, next addμL PEG300, mix and clarify, next addμL Tween 80, mix and clarify, next add μL saline, mix and clarify.

Method for preparing in vivo formulation: Take μL DMSO master liquid, next add μL Corn oil, mix and clarify.

Note: 1. Please make sure the liquid is clear before adding the next solvent.
2. Be sure to add the solvent(s) in order. You must ensure that the solution obtained, in the previous addition, is a clear solution before proceeding to add the next solvent. Physical methods such as vortex, ultrasound or hot water bath can be used to aid dissolving.
3. All of the above co-solvents are available for purchase on the GlpBio website.
