PolQi2 is a Polθ inhibitor that targets and inhibits alt-EJ (alternative end-joining) repair by inhibiting the helicase domain at the N-terminus of Polθ [1]. PolQi2 can target DNA repair pathways to promote homology-directed repair (HDR) by regulating DNA polymerase Polθ[2]. PolQi2 treatment, combined with AZD7648, elevated HDR levels and inhibited AZD7648-induced kilobase-scale deletions across multiple loci and cell types [3]. PolQi2, combined with DNA-dependent protein-kinase (DNA-PK) can reduce insertions and deletions (indels) and improve the accuracy of various leading editing systems[4].
