666-15 es un inhibidor selectivo de la proteína de unión al elemento de respuesta de AMP cíclico (CREB) con un valor de IC50 de 0.081±0.04μM[1]. 666-15 también inhibe la expresión de genes objetivo endógenos de CREB, a saber, el nivel de transcripción de la proteína relacionada con el receptor nuclear 1 (Nurr1/NR4A2)[1]. CREB es un factor de transcripción nuclear que puede ser activado por una variedad de señales extracelulares (incluidos factores de crecimiento y hormonas)[2]. 666-15 bloquea el efecto neuroprotector del inhibidor de necrosis necrotizing inhibitor 1 (nec-1), que es un inhibidor específico y efectivo de la necroptosis[3].
In vitro, el tratamiento de miocitos cardiacos de recién nacidos de rata (NRCMs) con 666-15 (1µM) durante 2 horas inhibió efectivamente la fosforilación de CREB inducida por fenilefrina, pero no tuvo un efecto significativo en la fosforilación de p38[4]. El tratamiento de condrocitos de cobayas envejecidas con 666-15 (0-500nM) redujo significativamente el nivel de proteína de p-CREB1 y aumentó la viabilidad celular[5]. El tratamiento con 666-15 (0-10µM) de células HEK293T transfectadas con diferentes factores de transcripción durante 5-7 horas mostró poca o ninguna inhibición de la transcripción génica mediada por Gal4-MLL, Gal4-c-Myb, Gal4-TEAD4/YAP1 y el factor de respuesta sérica (SRF)[6].
In vivo, 666-15 (10mg/kg) se inyectó intraperitonealmente en ratones trasplantados con células de cáncer de mama triple negativo (TNBC) e inhibió el crecimiento del cáncer de mama. La combinación con docetaxel (DOC) mostró mejores efectos y no tuvo un efecto significativo en el peso corporal de los ratones[7].

Después de que se formaron los tumores, los ratones se dividieron en cuatro grupos y se les inyectó intraperitonealmente con vehículo, docetaxel (DOC) (10mg/kg), 666-15 (10mg/kg) o ambos una vez a la semana. Se monitoreó el tamaño del tumor una vez a la semana y se tomaron imágenes del tumor después de sacrificar a los ratones. |
