BOLD-100 es un agente anticancerígeno a base de rutenio [1]. BOLD-100 tiene un alto potencial de focalización tumoral, se une fuertemente a proteínas séricas como la albúmina y la transferrina, y se activa en el entorno tumoral reductor [2]. BOLD-100 puede interrumpir la homeostasis del retículo endoplasmático (ER) e inducir estrés ER y respuesta de proteínas desplegadas (UPR)[3].
In vitro, el tratamiento BOLD-100 (100μM) de células MCF7(2) durante 72h redujo significativamente las células de fase S, aumentó las células de fase G2/M, redujo significativamente las proteínas del ciclo celular (como RAD51, PCNA, ATM), e indujo un aumento en las especies reactivas de oxígeno intracelular (ROS)[4]. El tratamiento BOLD-100 (100μM) de células HCT116 durante 24h mejoró la fosforilación de la serina eIF2A 51 en las células, pero redujo la expresión de la chaperona ER basal GRP78[5]. BOLD-100 (0-200μM) tiene un efecto inhibidor significativo en las líneas celulares malignas de varias fuentes, como cáncer de hígado, melanoma, cáncer de hígado, melanoma, cáncer de pulmón y cáncer de colon, cuando se trata durante 72h, con un valor de IC50 de 45-200μM[6].
In vivo, BOLD-100 (30mg/kg) se trató mediante inyección intravenosa durante 2 semanas en ratones con xenoinjertos de células Hep3B, lo que inhibió significativamente el crecimiento de tumores transplantados en ratones, mejoró la tasa de supervivencia de ratones y aumentó el número de células apoptóticas en los tejidos tumorales. Tiene un efecto terapéutico sinérgico con sorafenib [6]. BOLD-100 (50mg/kg) se trató por inyección intravenosa durante 3 semanas en ratones con xenoinjertos de células VACO432, pero el efecto inhibidor sobre el crecimiento tumoral no fue obvio. El tratamiento combinado con AZD6738 puede conducir a una ralentización en el crecimiento tumoral, pero la tolerancia es pobre y los ratones pierden peso[7].

Método de preparación | Las células se cultivaron a una confluencia del 70% en 100mm en medio de crecimiento completo durante 24h. Al día siguiente, las células fueron tratadas con vehículo o 100μM BOLD-100 durante 72h adicionales. Luego, las células se fijaron en etanol y se analizaron mediante la citometría de flujo. Cada experimento se repitió al menos tres veces. |
