Gap 26, como péptido mimético de connexina, que tiene las regiones de aminoácidos SHVR que contribuyen crucialmente al acoplamiento de conexiones para generar uniones de bracha [1]. Gap 26 puede inhibir la liberación de ATP inducida por un aumento en el calcio [2]. Gap 26 inhibió el acopllamiento eléctrico en pares de células mediados por uniones de brecha [3].
Gap 26 (25μM; 24h) atenuó la apoptosis de las células epiteliales tubulares[4]. Gap 26 (150μM; 24h) disminuyó la producción de ROS, inhibió la señalización ASK1-JNK/p38 y disminuyó la apoptosis en las células epiteliales alveolares de tipo II de ratas (RLE-6TN)[5].
Gap 26 (1μg/kg/día; i.p. ; 3 veces a la semana durante 8 semanas) alivió la hiporesponsividad cronotrópica y la gravedasd del daño hepático y tuvo un efecto antioxidante en el corazón en el modelo de ratas de cardiomiopatía cirrótica [6]. Gap 26 (25μg/kg; i.p. ; 4 día) deterioró aún más la destrucción de las sinapsis y disminuyó la densidad de la columna vertebral del hipocampo en el modelo de ratas de oclusión de la arteria cerebral media [7].

Método de preparación | Las ratas fueron anestesiadas con un 10% de chloral hydrate. Posteriormente, se hizo una incisión en la línea media en el cuello de las ratas. La arteria carótida común derecha (CCA), la arteria carótida externa (ECA), y la arteria carótida interna (ICA) fueron aisladas. La ECA se cortó aproximadamente 4mm por encima de la bifurcación común de la arteria carótida. Se insertó una sutura de monofilamento de nailon de 4.0-s en ICA a través del muñón hasta la resistencia. De esta manera, se bloqueó el flujo sanguíneo a la arteria cerebral media (MCA). 2 horas, después, se extrajo el filamento para la reperfusión y se ligó la incisión. Gap 26 se disolvió en solución salina y se administró por vía intraperitoneal a una dosis de 25 μg/kg una vez al día. Para evitar la influencia en la fase aguda, optamos por aplicar Gap 26 desde el día 3 después de la isquemia, cuando el volumen de infarto se estabilizó y comenzó el período de reparación. A los 7 días después del MCAO, se sacrificaron ratas y se extrajeron cerebros lo antes posible. Se aplicó Rapid Golgi Stain Kit para procedimientos de preparación y tinción de tejidos. Todos los procedimientos de tinción de Golgi-Cox se llevaron a cabo en estricta conformidad con las instrucciones. Usando un microtomo de congelación, cada bloque se cortó coronalmente en secciones en serie de 100 μm de espesor a partir de +1,00 a 5,00 mm del bregma. Las secciones se montaron en portaobjetos y se observaron bajo un microscopio confocal de barrido láser. |
