Description of ML-162

ML-162 es una molécula pequeña que inhibe Glutathione peroxidase 4 (GPX4), con un valor de IC50 de 1.42µM[1]. ML-162 suprime potwentemente la tioredoxina reductasa 1 (TXNRD1) con un valor de IC50 de 19.5µM[2]. ML-162 se uso mucho como compuesto modelo para explorar la unión covalente de ML-162 a los lugares activos y catalíticos de GPX4 y para desarrollar derivados relacionados[3].

In vitro, el tratamiento con ML-162 duranteor 48 horas inhibió significativamente la proliferación de células HT1080 con un valor de IC50 de 0.6±0.09μM[4]. El tratamiento de células HK-2 con 1μM ML-162 durante 24 horas inhibió significativamente la via durante 24h causó significativamente la muerte de las células Pfa1 y bloqueó la inhibición de la actividad de la ferroptosis de 5mM N-acetyl-l-cysteine (NAC) [6].

In vivo, el tratamiento con ML-162 a través de inyección intravenosa a una dosis de 20mg/kg/día durante 19 díays inhibió mucho el crecimiento del tumor en un modelo de ratón desnudo BALB/c xenoinjetado con el tumor HT1080, y disminuyo los niveles de GPX4 y Bcl-2 en los tejidos tumorales[4]. El tratamiento combinado con 1mg/kg de ML-162 y 10mg/kg de C7 a través de inyección intraperitoneal dos veces cada tres días durante 21 días inhibió mucho el crecimiento tumoral en el ratón del modelo de xenoinjerto HepG2 sin influir el peso corporal de los ratones[7]. La inyección intraperitoneal diaria de ML-162 a una dosis de 40mg/kg/día durante 2 semanas indujo mucho la ferroptosis tumoral en un modelo de ratón de cáncer de mama y resultó en un incremento de los niveles de PTGS2, MDA y 4-HNE en los tejidos tumorales[8].

Método de preparación

Las células HepG2 se cultivaron en DMEM (glucosa alta), suplementada con suero bovino fetal (FBS) al 10%, a 37℃ en una atmósfera humidificada que contiene 5% de CO2. Las células HepG2 se sembraron en placas de 96 pozos y se incubaron durante 24 horas. Las células fueron tratadas con el vehículo o ML-162 (100nM) durante 0, 6, 12, 18, 24h, respectivamente. Después, se añadieron 10µl de solución CCK-8 a cada pozo, y la absorbancia se midió a 450 nm.

Ratones desnudos BALB/c

Método de preparación

El modelo de xenoinjerto HT1080 se estableció mediante la inyección subcutánea de células HT1080 (4×106/ratón) en el abdomen derecho de ratones desnudos BALB/c en un entorno estándar. Cuando el tamaño del tumor era de aproximadamente 80-100 mm3, los ratones se dividieron aleatoriamente en cuatro grupos (5 ratones en cada grupo), incluido el grupo salino, ML-162 (20 mg/kg), GIC-20 (20 mg/kg) y GIC-20 (40 mg/kg). Los ratones fueron inyectados por vía intravenosa diariamente durante 19 días, y el volumen tumoral y el peso corporal se registraron cada dos días durante el tratamiento farmacológico. Después del tratamiento, los ratones fueron sacrificados sin dolor, y los tejidos tumorales fueron recogidos para hematoxilina-eosina (H&E) e inmunohistoquímica.
