ML351 es un inhibidor selectivo de 15-lipoxygenase-1 (15-LOX-1; 12/15-LOX) con un IC50 de 200nM[1]. 15-LOX-1 es un miembro de la familia de la oxidasa lipídica que puede desencadenar la peroxidación de fosdolípidos en la membrana plasmática. Al inhibir el 15-LOX-1, ML351 puede reducir la producción de productos oxidativos dañinos en las células[2].
In vitro, las células HT-22 tratadas con ML351 (0-10μM) inversamente la muerte celular inducida por glutamato de forma dependiente de la dosis[1]. El tratamiento ML351 (10μM) de células de macrófagos peritoneales (PMφ) durante 2 h inhibió la respuesta inflamatoria inducida por Kdo2-Lipid A y suprimió la expresión de mRNA de 12/15LOX y óxido nítrico sintasa inducible (iNOS )[3].
In vivo, ML351 (50 mg/kg) tratado con ratones con insuficiencia cardíaca aguda por inyección subcutánea redujo significativamente el tamaño del infarto de miocardio en los días 1 y 5, aumentó la expresión de CD11b y redujo las respuestas inflamatorias en el corazón y el bazo[3]. ML351 (24 mg/kg) tratado con inyección intraperitoneal en ratones diabéticos no obesos (NOD) durante 8 semanas redujo los niveles de glucosa en sangre, redujo los marcadores de estrés oxidativo en las células β pancreáticas y aumentó los niveles de enzimas antioxidantes[4]. ML351 (50μM/kg) tratado con inyección intraperitoneal en ratas sometidas a tratamiento de ortodoncia durante 14 días inhibió fuertwmente la resorción de raíces inducida por la ortodoncia (OIRR) y suprimió la aparición de osteoclas y odontostos [5].
