Patulin es una lactolactona polifenólica tóxica [1]. Patulin reacciona con los grupos de proteína sulfhidrilo, desencadenando el estrés oxidativo celular y el daño del DNA mediado por p53, lo que daña la barrera epitelial intestinal [2]. Patulin tiene propiedades teratogénmicas, genotóxicas y neurotóxicas [3-4].
En las células HL-60, después de Patulin (0-10μM; 6h) disminuyó significativamente la viabilidad celular[5]. En las células HepG2, Patulin (0.5-2μM; 24h) induce la autofagia en las células a través de la vía ROS-Akt1-MTOR [6].
En ratones CF-1, Patulin (1.0-3.75mg/kg; ip; inyección única) indujo rupturas de la cadena de DNA, disminuyó los niveles de GSH y aumentó la peroxidación lipídica en el cerebro, el hígado y el riñón [7]. En el modelo de ratón de xenoinjerto de células B16F10, Patulin (5-50μg/kg; ip; 20d) indujo significativamente la regresión tumoral [8].

Método de preparación | El diseño experimental involucró nueve grupos de 10 ratones, alojados individualmente en jaulas. Ratones recibieron una sola dosis intraperitoneal (i.p.) de 1,0, 2,5 o 3,75 mg/kg de peso corporal (BW) de Patulin. La solución de caldo de Patulin se disolvió en DMSO y luego se diluyó con solución salina hasta una concentración final de DMSO inferior al 0,1 %. Los ratones se dividieron en los siguientes grupos: Grupo 1: Grupo de control, administrado con 0,1% de DMSO en solución salina (5 ml/kg de peso corporal). Grupo 2: Grupo de control positivo, administrado con MMS (40 mg/kg de peso corporal). Grupos 3-5: La patulina se administró por vía intraperitoneal en las siguientes dosis: Grupo 3, 1,0 mg/kg de peso corporal; Grupo 4, 2,5 mg/kg de peso corporal; Grupo 5, 3,75 mg/kg de peso corporal. Grupo 6: Se administró N-ac (1840 μmol/kg de peso corporal) 3 horas antes de la administración de DMSO al 0,1 % (5 ml/kg de peso corporal). A los grupos 7-9 se les inyectó por vía intraperitoneal con Patulin en las siguientes dosis: 1,0 mg/kg de peso corporal para el Grupo 7; 2,5 mg/kg de peso corporal para el Grupo 8; y 3,75 mg/kg de peso corporal para el Grupo 9. Todos los animales de los grupos 7-9 recibieron 300 mg/kg de N-ac 3 horas antes de Patulin. |
