Hecubine is a natural aspidosperma-type alkaloid found in E. officinalis, with anti-neuroinflammation properties [1]. Hecubine can alleviate neuroinflammation and oxidative stress by modulating the Nrf2 and TLR4 signaling pathways [2]. Hecubine has been widely used to inhibit the growth of various types of cancer cells[3].
In vitro, Hecubine pre-treatment (25μM) for 1 hour can effectively reduce the production of NO and PGE2 and the expression of iNOS and COX-2 in BV2 cells stimulated by Lipopolysaccharide (LPS) [4].

Preparation Method | BV2 cells were cultivated in 24-well plate at a density of 4×105 cells/ml in DMEM medium supplemented with 10% fetal bovine serum (FBS), phosphate-buffered saline (PBS), and 1% penicillin/streptomycin. Cells were treated with different concentrations of the Hecubine (0, 6, 12, and 25µM) for 1h prior to LPS stimulation (800ng/ml). The levels of NO, PGE2, and IL-6 in cells were determined. |
