A-803467 est un bloqueur du canal sodique NaV1.8 résistant àà la tétrodotoxine; il bloque le canal NaV1.8 humain avec IC50 de 8nM[1, 2]. A-803467 peut efficacement la douleur inflammatoire et neuropathique expérimentale induite par le canal NaV1.8[1].
A-803467 (7.5μM; 2h) a augmenté l'accumulation intracellulaire de [3H]-MX dans les cellules HEK293 ABCG2-transfectées[2]; A-803467 (7.5μM; 0, 30, 60, 120min) a significativement bloqué l'efflux intracellulaire de [3H]-MX depuis des cellules transfectées avec ABCG2 [2]; A-803467 (100nM) a induit un blocage significatif du courant dans les cellules HEK-293 exprimant le canal sodique NaV1.8 humain[1]; A-803467 (30nM) a raccourci le potentiel d'action dans les myocytes ventriculaires de rats stimulés à basse fréquence (0,1Hz)[3]; A-803467 (30nM; 15min) a réduit la fréquence des ondes de Ca2+ diastolique dans les cardiomyocytes auriculaires humains provenant de patients en rythme sinusal[4].
A-803467 (10, 30, 100mg/kg; i.p.) a réduit fortement l'hyperalgésie thermique dans le modèle de douleur inflammatoire chez le rat Sprague-Dawley (SD) traité à l'adjuvant complet de Freund (CFA)[1]; A-803467 (10, 30, 100mg/kg; i.p.) a réduit l'allodynie mécanique dans le modèle de lésion nerveuse spinale L5/L6 chez le rat SD[1]; A-803467 (10, 30, 100mg/kg; i.p.) a réduit la douleur articulaire chez les rats Wistar mâles induite par le monoiodoacétate (MIA)[5].
