AR7 est un antagoniste atypique du récepteur α de l'acide rétinoïque (RARα)[1]. AR7 antagonise le RARα pour lever son inhibition de l'ACM,augmentant ainsi significativement les niveaux de LAMP2A et l'activité de l'ACM, et favorisant la dégradation sélective des protéines mal repliées ou agrégées[2-3]. AR7 est couramment utilisé dans le traitement de la maladie de Parkinson [4].
Dans les neurones corticaux primaires, AR7 (0-20μM; 21j) atténue l'accumulation progressive d'oligomères de SNCA intracellulaires dans les cultures à long terme de neurones corticaux [5]. Dans les cellules SN4741, AR7 (20μM; 24h) réduit l'accumulation de gouttelettes lipidiques dans le modèle déficient en Wdr45 en activant l'autophagie médiée par les champeronnes (ACM) et en favorisant la dégradation de la Fasn, une enzyme de la biosynthèse lipidique [6].
Chez les souris C57BL/6, AR7 (10mg/kg; ip; 6h) atténue la stéatose hépatique induite par le diclofénac [7]. Chez les souris descendantes hétérozygotes, l'administration d'AR7 (10mg/kg; ip; 6 semaines) favorise l'élimination hépatique de SERPINA1E342K/ATZ et réduit la pathologie d'agrégation hépatique de SERPINA1E342K/ATZ [8].

Méthode de préparation | Afin de déterminer si le traitement par AR7 module les niveaux d'oligomères de SNCA, des neurones corticaux primaires matures (JIV9) ont été cultivés dans des conditions normales avec ou sans AR7 (0, 10 ou 20μM)pendant jusqu'à 21 jours. AR7 dissous dans DMSO de qualité pour culture cellulaire a été ajouté au milieu de culture aux JIV9 et DIV14 aux concentrations indiquées. Le milieu conditionné et les lysats cellulaires totaux ont été extraits aux DIV9, 14 et 21 pour analyse par Western blot et par ELISA des oligomères de SNCA. |

Méthode de préparation | Des souris mâles C57BL/6 (âgées de 7 semaines) ont été hébergées dans une pièce climatisée (24℃) avec un cycle jour/nuit de 12 heures et ont eu un accès libre à l'eau et à la nourriture pendant une semaine. Après une période d'acclimatation d'une semaine, les animaux ont été répartis au hasard en quatre groupes (n=6 souris chacun): Groupe 1, véhicule; Groupe 2, diclofénac; Groupe 3, AR7; et Groupe 4, diclofénac + AR7. Le groupe véhicule a reçu une injection intrapéritonéale de 0.5% de DMSO dans une solution saline. Le groupe diclofénac a reçu du diclofénac (100mg/kg, voie intrapéritonéale) deux fois toutes les 12 heures. Le groupe AR7 a reçu de l' AR7 (10mg/kg, voie intrapéritonéale) une heure avant le diclofénac. Le lendemain, les souris ont été anesthésiées avec Zolettiline 50 et sacrifiées. |
