AR7 es un antagonista atípico del receptor de ácido retinoico α (RARα) [1]. AR7 antagoniza RARα para aliviar su inhibición de la CMA, mejorando así significativamente la actividad de LAMP2A y CMA y promoviendo la degradación selectiva de proteínas mal plegadas o agregadas [2-3]. AR7 se usa comúnmente para tratar la enfermedad de Parkinson[4].
En las neuronas corticales primarias, AR7 (0-20μM; 21d) atenúa la acumulación progresiva de oligómeros SNCA intracelulares y extracelulares en cultivos a largo plazo de neuronas corticales [5]. En células SN4741, AR7 (20μM; 24h) reduce la acumulación de gotas lipídicas en el modelo deficiente en Wdr45 al activar la autofagia mediada por chaperona (CMA) y promover la degradación de la enzima de biosíntesis lipídica Fasn [6].
En ratones C57BL/6 , AR7 (10mg/kg; ip; 6h) alivia la esteatosis hepática inducida por diclofenaco [7]. En ratones descendencia heterocigota, la administración de AR7 (10mg/kg; ip; 6 semanas) promueve la eliminación hepática de SERPINA1E342K/ATZ y mitiga la patología de agregación hepática de SERPINA1E342K/ATZ [8].

Método de preparación | Para investigar si el tratamiento con AR7 modula los niveles de oligómero SNCA, las neuronas corticales primarias maduras (DIV9) se cultivaron en condiciones normales con o sin AR7 (0, 10 o 20μM) durante un másta de 21 días. AR7 disuelto en DMSO de grado de cultivo celular se añadió al medio de cultivo a DIV9 y DIV14 a las concentraciones indicadas. Se extrajeron los lisados medios y celulares totales acondicionados en DIV9, 14 y 21 para Western blotting y análisis ELISA de oligomero SNCA. |

Método de preparación | Los ratones machos C57BL/6 (7 semanas de edad) fueron alojados en una habitación con aire acondicionado (24℃) con un ciclo día/noche de 12 horas y se les permitió el acceso gratuito al agua y la comida durante una semana. Después de un período de aclimatación de una semana, los animales se dividieron aleatoriamente en cuatro grupos (n=6 ratones cada uno): Grupo 1, vehículo; Grupo 2, diclofenaco; Grupo 3, AR7; y Grupo 4, diclofenaco + AR7. El grupo de vehículos recibió una inyección intraperitoneal de DMSO al 0,5% en solución salina. El grupo de diclofenaco recibió diclofenaco (100 mg/kg, por vía intraperitoneal) dos veces cada 12 horas. El grupo AR7 recibió AR7 (10 mg/kg, por vía intraperitoneal) una hora antes del diclofenaco. Al día siguiente, los ratones fueron anestesiados con Zolettilina 50 y sacrificados. |
