Cholesterol esterase est une hydrolase qui catalyse la conversion du cholestérol ester ingéré en cholestérol libre et en acide gras libre[1]. Cholesterol esterase présente des propriétés pro-athérogènes par la facilitation de la conversion des particules lipoprotéiques plus grandes et moins athérogènes en sous-espèces lipoprotéiques plus petites et plus athérogènes [2]. Cholesterol esterase peut être immobilisée de manière covalente sur un film de polyaniline pour développer un biocapteur pour la mesure du cholestérol[3].
In vitro, un traitement par Cholesterol esterase (100mU/ml) durant 3 heures favorise la prolifération et la migration des cellules endothéliales de la veine ombilicale humaine (HUVEC) [4].

Lignées cellulaires | Des cellules endothéliales de la veine ombilicale humaine (HUVEC) |
Méthode de préparation | Des HUVEC ont été incubées dans du milieu EBM-2 additionné de 2% de sérum de veau foetal (SVF) dans une atmosphère humidifiée à 37°C avec 5% de CO2. La migration des HUVEC a été déterminée à l'aide d'une chambre chémotaxique de 24 puits avec un filtre en polycarbonate (taille des pores de 8-μm). Des HUVEC (2×104 cellules) dans un milieu sans sérum (100μl) ont été déposées dans le compartiment supérieur de la chambre. Le milieu contenant Cholesterol esterase (100mU/ml) a été ajouté dans le compartiement inférieur de la chambre et les cellules ont été incubées(37°C; 3 heures). Après le retrait de Cholesterol esterase et une incubation supplémentaire (37°C; 24 heures), les cellules non invasives sur la surface supérieure du filtre ont été complètement essuyées avec un écouvillon en coton et les filtres ont été fixés (éthanol à 70% dans PBS) et colorés (hématoxyline). Le nombre de cellules ayant migré a été compté dans 10 champs aléatoires à un grossissement de 200× au microscope. |
